The present experimental work was conducted to elucidate the biochemical changes in broiler chicken after ziram intoxication. For the study, 150 apparently healthy broiler chicks were reared together up to 2 weeks of age and thereafter allocated randomly to four groups, that is, control (n=30), acute (n=10), subacute (n=20) and subchronic (n=90). Toxicity was induced following single oral dose of Ziram at 100 mg/kg body weight for acute intoxication, 5 mg/kg body weight per day for 20 days in case of subacute group and 1 mg/kg body weight per day for the study period of 90 days in case subchronic intoxication. The mean values of liver function test viz. serum glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), serum alkaline phosphatase (ALP), serum lactate dehydrogenase (LDH) and bilirubin in acute intoxicated birds at 24 h post intoxication, subacute and subchronic groups were significantly (p<0.05) higher when compared with the control group. Further in kidney function test, a significant (p<0.05) increase in the mean values of blood urea nitrogen (BUN) was recorded in acute ziram intoxicated birds at 24 h interval and subchronic groups but no significant (p>0.05) effect was observed in the subacute group when compared with the control. The creatinine levels increased significantly (p<0.05) in all the intoxicated birds compared to control. Moreover, a significantly (p<0.05) higher increase in the zinc levels was observed in all the intoxicated birds compared with the control. The mean serum copper and calcium levels were significantly (p<0.05) lower in all the intoxicated birds compared to the control. In conclusion, the results were suggestive of the fact that ziram caused hepatotoxicity and nephrotoxicity in broiler chicken.

To evaluate the histopathological, immunohistochemical, and biochemical effects of liver changes after mancozeb administration.Methods: Rats were divided into groups-the control group (n=7) and the mancozeb group (n=7)-, given 500 mg/kg mancozeb dissolved in corn oil daily for four weeks by an orogastric tube. Caspase-3 and tumor necrosis factor-alpha (TNF-α) primary antibodies were used for immunohistochemical analysis. Results: Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) values of the mancozeb group increased significantly than ones of the control group. Venous dilatation, inflammation, hepatocyte degeneration, TNF-α, and caspase-3 expression scores increased significantly in the mancozeb group. In the mancozeb group, intensive caspase-3 expression was observed in hepatocyte cells around the central vein in the center of the liver lobule, and there was an increase in TNF-α expression in the inflammatory cells around the enlarged central vein and Kupffer cells and apoptotic hepatocyte cells. Conclusion: Subacute mancozeb exposure in rats leads to elevated toxicity with impaired liver function, increased inflammation in tissue and increased apoptosis due to cellular damage in the liver, and decreased liver regeneration ability due to congestion and degeneration of blood vessels.

Background: Metalaxyl is a fungicide used to control soil-borne fungal diseases on fruits, cotton, soy bean, peanuts and grasses. Food contamination with metalaxyl is a worldwide problem especially in developing countries. Ginger has antioxidant properties. Aim of the work: To elucidate the histological alterations that may occur in the liver tissue of rat by metalaxyl and to investigate the role of ginger supplementation against these alterations. Materials and methods: Thirty healthy adult male albino rats were used in this study. They were equally divided into three groups: Group I served as control group. Group II (metalaxyl-treated) received 130 mg/kg body weight/day of metalaxyl dissolved in distilled water using a gastric tube for 3 times/week for continuous 4weeks. Group III (metalaxyl-ginger treated group) received metalaxyl as group II in addition to ginger given orally at a dose of 100 mg/kg three times/week for successive 4 weeks. At the end of experiment, all rats were anaesthetized and liver specimens were taken and processed for light and electron microscope examination. Immunohistochemical staining was carried out for detection of the proapoptotic marker Bcl-2 antagonist-X (BAX) protein in hepatocytes. Area % of BAX protein was measured and statistically analyzed. Results: Examination of the liver sections of metalaxyl-treated group ІІ showed different degrees of focal lobular affection. Central veins and blood sinusoids were dilated and congested. The hepatic lobules lost their normal architecture and the hepatocytes showed cytoplasmic vacuolization with darkly stained nuclei. Enlarged portal areas with numerous bile ductules and cellular infiltration were detected. Ultrastrcturally, hepatocytes had condensed heterochromatin nuclei and electron lucent areas were observed in their cytoplasm. Significant increase in area % of positive immunoreaction for BAX was detected in comparison with control group and group ІІІ. Examination of liver sections of group ІІІ revealed preservation of nearly normal histological structure with slight congested blood vessels and few cellular infiltration. Conclusion: Metalaxyl had pathological effects on the structure of rat's liver and these effects were decreased by ginger.
