Effect of Lutein on Cytochrome P450 (Isoform CYP3A4) - An in vitro Study

Abstract: Background: Lutein is a carotenoids vitamin rich in many fruits and vegetables and also available in multivitamin products. It is consumed for its effect on eye disease, cancer, diabetes and other health conditions. Recently, herbal preparations are increasingly used in healthcare systems and concomitant administration of synthetic medications may cause pharmacokinetic or pharmacodynamic interactions leading to very serious medical problems. Understanding the ability of herbal extracts and preparations to modu… Show more

“…Prathapan et al used the same method for studying the effect of lutein on differentiation and the cells were incubated at different concentrations later quantified using multiple plate readers at 490 nm. This preparation was similar to the preparation used in the present study [17,18]. The leaves of coriander show stronger antioxidant activity than seeds, but in both the parts of coriander, ethyl acetate extract showed the strongest ability.…”

“…Of specific interest in this context is the CYP3A4 which is an important enzyme in the metabolism of drugs and several other xenobiotics. Interactions of xenobiotics including drugs with CYP3A4 have been demonstrated to either be inhibitory or inductive (Meenapriya et al., 2018), and such effect could influence the pharmacokinetics of any co‐administered agent. In this study, the absence of inhibitory effect of GLH on CYP3A4 could suggest that it is not likely to have inhibitory DDI with drugs currently metabolized by CYP3A4 isoform.…”

“…They have also been identified as major detoxification system in drug metabolism processes. Alterations in drug metabolism facilitated by other co‐administered agents or drugs could potentiate metabolic and/or systemic drug–drug interaction (DDI) (Meenapriya et al., 2018). Unpredicted DDIs have been implicated in severe adverse effects or treatment failures and usually involve either the induction or inhibition of specific CYP activity (Asha & Vidyavathi, 2010; Clarke & Jones, 2008; Meenapriya et al., 2018).…”

“…Since human cancer cell lines maintain hallmark characteristics of cancer cells, are easy to propagate, and can be genetically engineered to provide reproducible This procedure may also differentiate between cells in various stages of apoptosis. As a result, AO/EtBr staining may be used in DSTs [31]. The morphological changes in cells treated with Lippia nodiflora leaf extract were also detected, and the findings were significant.…”
