Effect of Low‐dose Irradiation on Pregnant Mouse Haemopoiesis

Abstract: Summary. The effects of low‐dose gamma radiation to haemopoietic progenitor cell compartments of the marrow and spleen of virgin female mice and pregnant mice were studied. Microplasma clot cultures were used to assess burst‐forming uniterythroid (BFU‐E) and colony‐forming unit‐erythroid (CFU‐E) activity, and double‐layer agar cultures were established to evaluate granulocyte‐macrophage colony‐forming cell (GM‐CFC) and macrophage colony‐forming cell (M‐CFC). The apparent shift in maternal erythropoiesis from t… Show more

“…PBMCs were purified from venous blood (Ͻ3.0 ml; n ϭ 37) collected in EDTA-containing vials using Ficoll/Hypaque as described previously (45). Cells were plated at 1 ϫ 10 6 cells per ml in Dulbecco's modified Eagle's medium (DMEM) containing 10% pooled human serum (heat inactivated at 56°C for 30 min) and cultured for 48 h. SCGF genotyping.…”

“…PBMCs were purified from venous blood (Ͻ3.0 ml from Kenyan children [n ϭ 55] and 40 ml from malaria-naïve U.S. adults [n ϭ 11]) by use of Ficoll-Hypaque as described previously (48). Cells were plated at 1 ϫ 10 6 cells per ml in Dulbecco's modified Eagle's medium containing 10% pooled human serum (heat inactivated at 56°C for 30 min).…”
