Effect of long-term treatment with classical neuroleptics on NPQ/spexin, kisspeptin and POMC mRNA expression in the male rat amygdala

Abstract: Neuroleptics modulate the expression level of some regulatory neuropeptides in the brain. However, if these therapeutics influence the peptidergic circuits in the amygdala remains unclear. This study specifies the impact profile of the classical antipsychotic drugs on mRNA expression of the spexin/NPQ, kisspeptin-1 and POMC in the rat amygdala. Animals were treated with haloperidol and chlorpromazine for 28 days prior to transcript quantification via qPCR. Haloperidol and chlorpromazine induced a change in the… Show more

“…It is worth noting here that the physiological effect of olanzapine observed in the brainstem turned out to be identical to that of escitalopram (an antidepressant) in the hypothalamus. Our results, however, do not correspond with increased NPQ/spexin mRNA expression in the amygdala of rats exposed to the classic antipsychotic drug chlorpromazine [46].…”

“…Potential effects of antidepressants on the kisspeptin signalling is so far unveiled and there are no articles available dealing with the relations between SSRIs and kisspeptin/Kiss1R expressions in the brain. Our data are only approximate to the study for kisspeptin after chronically administered classical neuroleptics haloperidol and chlorpromazine, where a decrease Kiss-1 expression was noted in the amygdala [ 15 ]. Kisspeptin facilitates hippocampal synaptic transmission by activation of MAP kinase (MAPK) pathway in the granular cells of dentate gyrus.…”

“…found that chronic i.p. administration with haloperidol and chlorpromazine, the classical antipsychotic drugs, increased the spexin and pomc mRNA levels while decreased the kisspeptin-1 mRNA level in the rat amygdala (Palasz et al., 2018). It indicates that the neuropeptide spexin in the amygdala may be related to the mechanism of the antipsychotic drugs.…”
