Effect of Kelulut Honey on the Cellular Dynamics of TGFβ-Induced Epithelial to Mesenchymal Transition in Primary Human Keratinocytes

Abstract: Over-induction of epithelial to mesenchymal transition (EMT) by tumor growth factor beta (TGFβ) in keratinocytes is a key feature in keloid scar. The present work seeks to investigate the effect of Kelulut honey (KH) on TGFβ-induced EMT in human primary keratinocytes. Image analysis of the real time observation of TGFβ-induced keratinocytes revealed a faster wound closure and individual migration velocity compared to the untreated control. TGFβ-induced keratinocytes also have reduced circularity and display a … Show more

“…Keloid scar formation is due to the excessive induction of EMT by TGFβ in keratinocytes. In a study, treatment of 0.0015% Kelulut honey has been demonstrated to reduce the TGFβ-induced EMT in human primary keratinocytes, indicating Kelulut honey’s therapeutic potential in preventing keloid scar formation [ 132 ]. Rats pretreated with Kelulut honey showed significantly reduced total area of ulcer and ulcer index compared with an untreated ethanol-induced gastric ulcer rat [ 133 ].…”

“…However, in recent years, more and more studies have revealed that epidermal keratinocytes also play an important role in wound healing and scar formation [2,3]. During scar formation, keratinocytes gradually lose their characteristics of epithelial cells and acquire features of mesenchymal cells, manifested by decreased ability of cell adhesion and increased ability of migration [4]. However, it is still largely unknown that AGING how is the epithelial-mesenchymal transition (EMT) process regulated in dermal keratinocytes.…”

“…Davison SP indicated that epidermal‐mesenchymal interaction might play a key role in the pathogenesis of keloid, and further suggested combination therapy appears to be the most efficacious and safest keloid management option available 31 . It is the latest research trend to apply epithelial‐to‐mesenchymal transition to coordinate the complex pathway mechanism of keloid and to view the occurrence and development of keloid from a dynamic perspective 32,33 . TGF‐β is the most critical growth factor in keloid formation.…”
