Laboratory-produced alginate-bead inoculum of ectomycorrhizal (ECM) fungi Cortinarius sp. and Gomphidius glutinosus, fungal commercial products ECtovit® and Trichomil®, bacterial commercial product BactoFil B®, and commercial rooting stimulator Vetozen® were applied to a peat-perlite (1:2, v:v) rooting substrate of Norway spruce stem cuttings collected from 4-year-old nursery-grown seedlings immediately before the insertion of cuttings into the substrate. The application of beads free of fungi and the substrate without an additive were the other treatments. The cuttings were rooted in vessels (60 cuttings per vessel, 180 per treatment) placed in a glasshouse and arranged in a randomized complete block design. The cuttings were estimated for one growing season (approximately for 26 weeks) after their insertion into the rooting substrate. Rooting % of the cuttings ranged from 45 (mycelium-free beads) to 75 (control) according to treatments, 64 on average. No significant differences among treatments were found in % of ECM morphotypes, total ECM colonization of roots (%), and growth parameters of shoots and roots of the cuttings. The applied microbial additives were not sufficiently efficient to form treatment-related ectomycorrhizas that were formed by naturally occurring ECM fungi. Inoculation by the ECM fungus Cortinarius sp. and application of Trichomil had a partial stimulative effect on the shoot growth of cuttings. Shoot and root growth parameters were not significantly correlated with total ECM colonization, except for a negative dependence of the root number in Trichomil treatment. A higher concentration of K but lower concentrations of Ca and Mg in Ectovit treatment than in the other treatments were detected.

ResumenDesde el trabajo publicado por Pardos en 1962, se ha desarrollado una importante actividad investigadora en el campo de las ectomicorrizas en España; especialmente en los últimos 20 años. En este artículo, revisaremos los avances conseguidos en el estudio de los hongos ectomicorrícicos en lo que se refiere a su aplicación a la producción de planta para repoblación y a los resultados obtenidos en plantaciones experimentales. Los resultados publicados, principalmente con coníferas inoculadas con hongos de los géneros: Hebeloma, Laccaria, Pisolithus, Rhizopogon, Scleroderma o Suillus, demuestran que la micorrización controlada con hongos seleccionados mejora los procesos de repoblación y revegetación en distintas situaciones ambientales, como: el establecimiento de masas en suelos forestales productivos, tanto con especies autóctonas como con especies exóticas de rápido crecimiento, la repoblación de suelos agrarios abandonados, o la revegetación de suelos áridos de la zona mediterránea.Palabras clave: ectomicorriza, vivero forestal, micorrización de plantones, supervivencia y crecimiento en plantación, reforestación, revegetación. Abstract Controlled inoculation of reforestation seedlings with ectomycorrhizal fungi: current knowledge in SpainAfter the paper published by Pardos in 1962 a considerable effort of investigation on ectomycorrhizas has been done in Spain, especially during the last 20 years. In our article we review the progress achieved in our country concerning the knowledge of ectomycorrhizal fungi, in particular their use in the controlled inoculation of nursery grown plants for reforestation and revegetation purposes, and the performance of these mycorrhizal plants in experimental plantations. Published results mainly involve different conifer species inoculated with fungi belonging to the genera: Hebeloma, Laccaria, Pisolithus, Rhizopogon, Scleroderma or Suillus. These results show that controlled mycorrhizal inoculations with selected fungi improve the reforestation and revegetation processes in defined environmental situations, such as: reforestation of productive forest soils, both using native or introduced tree species, reforestation of set-aside agricultural lands, or revegetation of arid soils in the Mediterranean regions of Spain.Key words: ectomycorrhiza, forest nursery, seedling mycorrhization, field performance, reforestation, revegetation. Acidification of forest ecosystems leads to increased plant availability of the micronutrient manganese (Mn), which is toxic when taken up in excess. To investigate whether ectomycorrhizas protect against excessive Mn by improving plant growth and nutrition or by retention of excess Mn in the hyphal mantle, seedlings of two populations of Douglas fir (Pseudotsuga menziesii), two varieties, one being menziesii (DFM) and the other being glauca (DFG), were inoculated with the ectomycorrhizal fungus Rhizopogon subareolatus in sand cultures. Five months after inoculation, half of the inoculated and non-inoculated seedlings were exposed to excess Mn in the nutrient solution for further 5 months. At the end of this period, plant productivity, nutrient concentrations, Mn uptake and subcellular compartmentalisation were evaluated. Non-inoculated, non-stressed DFM plants produced about 2.5 times more biomass than similarly treated DFG. Excess Mn in the nutrient solution led to high accumulation of Mn in needles and roots but only to marginal loss in biomass. Colonisation with R. subareolatus slightly suppressed DFM growth but strongly reduced that of DFG (−50%) despite positive effects of mycorrhizas on plant phosphorus nutrition. Growth reductions of inoculated Douglas fir seedlings were unexpected since the degree of mycorrhization was not high, i.e. ca. 30% in DFM and 8% in DFG. Accumulation of high Mn was not prevented in inoculated seedlings. The hyphal mantle of mycorrhizal root tips accumulated divalent cations such as Ca, but not Mn, thus not providing a barrier against excessive Mn uptake into the plants associated with R. subareolatus.
