The present study was conducted to observe the effect of osmolality of glycerolated TEST-yolk glycerol extenders on post-thawing sperm kinematics of ram spermatozoa of the native Malpura breed maintained in a semi-arid tropical environment. Good quality semen obtained from adult rams was pooled, split and diluted to 1,000 million spermatozoa per ml in complete TEST-yolkglycerol extenders of 900, 1,200, 1,500 and 1,800 mOsm/kg osmolality. Diluted semen samples were loaded in 0.25 ml straws and cooled down to -125°C freezing temperature at the rate of -25°C per minute under controlled conditions before plunging into liquid nitrogen for storage. The thawing of straws was performed at 50°C in a water bath for 10 seconds and sperm kinematics of the frozenthawed spermatozoa were assessed by a computer-assisted sperm analysis technique. Osmolality of diluent had no significant effect on post-thawing % motility, % rapid, % medium and % slow moving frozen-thawed spermatozoa but significantly (p< 0.05) affected the % linearity and % straightness.The post-thawing % motility and % rapid motile spermatozoa were highest in samples extended in diluent of 1,500 mOsm/kg osmolality and lowest in 900 mOsm/kg. The curvilinear velocity of spermatozoa was significantly (p<0.05) higher for samples extended in 1,800 mOsm/kg, compared to those in 900 and 1,200 mOsm/kg, but the effect was not significantly different to those extended in diluent of 1,500 mOsm/kg osmolality. The study indicated that ram spermatozoa could tolerate a wide osmolality range for dilution in the complete TEST-yolk-glycerol extender for their cryosurvival. The highest recovery of motile spermatozoa following thawing was achieved in samples extended in the TEST-yolk-glycerol diluent of 1,500 mOsm/kg osmolality.

Goats are important livestock species of India. Chegu is a pashmina producing goat native to the cold arid region of Himachal Pradesh (H.P.), India. Semen cryopreservation from six Chegu elite males (aged 2.05±0.40 years; weighed 29.16±2.02 kg) was practiced using Tris Citrate Egg Yolk extender containing 10% EY and 6% Glycerol. Gross semen parameters includes volume (0.80.85±0.07 ml), color (Creamy white to yellowish), concentration (2238.5±231.0 x106 spermatozoa/ml) and mass motility (3.92±0.03).The significant changes (P less than 0.01) in post thaw seminal parameters (75.48±0.69 v/s 37.38±0.90; progressive motility), viability (75.79±0.95 v/s 48.25±1.78), morphological abnormalities (5.64±0.29 v/s 7.02±0.32) and HOST reactive spermatozoa (64.07±1.75 v/s 43.35±1.79) were observed in present study. Artificial insemination using frozen thawed semen having concentration (150 x106 spermatozoa/straw) from three different bucks was practiced in 40 synchronized goats with conception rate of 42.5 per cent. Non-significant variations amongst different bucks were observed with birth of 1.12 kids per doe and twinning rate of 11.8 per cent. It was concluded that semen cryopreservation along with artificial insemination can be practiced in Chegu goats to improve the population of this endangered species.

After thawing, the initial freezing temperature has a considerable impact on spermatozoa motility and velocity; however, the best motility of spermatozoa may be attained at −125°C [ 135 ]. Germplasm storage and transportation in artificial insemination (AI) and other advanced technologies are facilitated by cryopreservation. In reproduction, the cryopreservation of sperm allows it to be transported across vast distances and used even after the sire’s death. However, the technique of cryopreservation might damage sperm and limit their activity. Several cryobiological investigations have reported that the integrity of the sperm membrane is frequently involved in the physical and biological elements that affect sperm survival at low temperatures during the cryopreservation process. However, successful cryopreservation of ram sperm is still a work in progress because a considerable percentage of sperm do not survive the freezing and thawing process. Sperms are destroyed during cryopreservation of semen due to varying concentrations of cryoprotective chemicals and if semen is not cooled at optimal cooling rates. Hence, it is crucial to know the optimum cooling rates with freezing and thawing protocols for maximum recovery of viable and functional sperm cells for a successful cryo-freezing of ram spermatozoa. Therefore, the current study compiled and compared the research on the impact of different cryopreservation procedures, cooling rates, equilibration time, and thawing protocols on post-thaw ram semen quality.
