Effect of indomethacin and prostaglandin E2 on structural differentiation of rat endometrium during artificially induced decidualization

Abstract: Morphological differentiation of uterine stromal and luminal epithelial cells was studied in steroid-injected ovariectomized rats following unilateral intrauterine instillation of sesame oil, phosphate-buffered saline containing gelatin (PBSG), PBSG + indomethacin (IM; an inhibitor of prostaglandin synthesis), or PBSG + IM + prostaglandin E2 (PGE2). The latter two treatments were preceded by a subcutaneous injection of IM. Uteri were examined by light and electron microscopy at intervals between 8 and 120 hr (… Show more

“…We also found that the size of the implantation sites and decidualization were severely affected by nimesulide, although nimesulide had no effect on the number of implantation sites (J. Cong et al, unpublished data). When infused into the lumen of sensitized rat uteri in which endogenous PG synthesis is inhibited, PGE2 is able to restore the endometrial vascular permeability response and decidualization (Kennedy & Lukash 1982, Keys & Kennedy 1990, Hamilton & Kennedy 1994. In steroid-injected ovariectomized rats, following unilateral intrauterine instillation of sesame oil, administration of indomethacin inhibited the onset of the decidual cell reaction and had deleterious effects on the luminal epithelium.…”

“…In steroid-injected ovariectomized rats, following unilateral intrauterine instillation of sesame oil, administration of indomethacin inhibited the onset of the decidual cell reaction and had deleterious effects on the luminal epithelium. Inclusion of PGE2 in the instillate accelerated stromal cell differentiation and overcame the inhibitory effect of indomethacin (Keys & Kennedy 1990). All of these data suggest that PGE2 is essential for rat decidualization.…”

“…En revanche, l'influence de ces molécules sur la décidualisation endométriale est démon-trée dans d'autres espèces [16]. Les Pg sont impliquées dans l'ouverture de la fenêtre implantatoire dans l'espèce humaine.…”

“…Concentration of PGE, PG F and PGI is elevated in implan tation sites [57,62,71,72] and in the uteri of pseudopregnant animals induced to decidualize [60, 61,73], PG F2tt was shown to induce im plantation when administered systemically in the mouse [69], ham ster [42] and rat [74], On the other hand, PGE2 is of equal or greater potency for inducing decidualization when injected or infused intraluminally [58,70,75] and stable analogues of PGE2, but not of PG F2a, were able to induce decidualization in the rat [76], In addition, in spayed, steroidtreated rats induced to decidualize, the mor phological onset of decidualization is inhibit ed by indomethacin. Inclusion of PGE2 in the instillate overcame the inhibition [77].…”
