Effect of Heparin and Heparin Fractions on Platelet Aggregation

Abstract: A B S T R A C T Porcine intestinal mucosal heparin induced aggregation of platelets in citrated platelet-rich plasma and enhanced platelet aggregation and serotonin secretion induced by other agents. This action ofheparin was blocked by substances that elevate platelet cyclic AMP and by EDTA but not by inhibitors of platelet cyclooxygenase. The effect was not inhibited by apyrase or by N-amylthio-5'-AMP and therefore did not require the action of ADP, nor was there activation of platelet phospholipase. Platele… Show more

“…Thrombocytopenia of this type is mild (platelet count, >100 x 10 9 cells/L), not progressive, nor associated with bleeding or thrombosis (Salzman et al, 1980), and is independent of any immune reaction (Chong et al, 1993a;Burgess & Chong, 1997;Shantsila et al, 2009). It is characterized by a transitory, slight and asymptomatic reduction in platelet count, occurring during the first 1-2 days of heparin administration.…”

“…Heparin binds to platelets, activating platelet factor IV and stimulating its release (Salzman et al, 1980). Then, heparin creates a complex with platelet factor IV, stimulating antibodies primarily responsible for thrombocytopenia (Kelton et al, 1994).…”

“…Danaparoid is also known to have potent anticoagulant activity similar to that of heparin, [4], [5] although platelet aggregation was in some cases observed in blood with heparin [6]- [8] due to unclarified mechanism.…”

“…[9] Danaparoid would be then a promising antico- control. [8] PRP was pre-incubated for one minute in the incubating hole of the aggregometer. Automated blood counting Heparin (22.5 U) or Danaparoid (22.5 U) was placed in plastic test tubes as an anticoagulant, and 1 mL of whole blood was added to the tubes in duplicate.…”
