Effect of growth factors on matrix synthesis by human nasal chondrocytes cultured in monolayer and in agar

Abstract: Reconstructive surgery of multiple areas of the body may require replacement bone or cartilage transplants to repair defects or lesions of skeletal tissue. Advances in cell and tissue culture techniques now permit synthesis of autologous human cartilage in vitro. Several growth factors regulate the metabolism and activation of cartilage cells. To enhance culture conditions and effectiveness for in vitro cartilage engineering, the aim of our investigations was to characterize the influence of transforming growt… Show more

“…Members of the TGF-β superfamily play a major role in cartilage development and repair. Mainly, the isoforms TGF-β1, 2 and 3 enhance chondrocyte proliferation and increase ECM synthesis by chondrocytes (Morales 1991; Bujia et al 1996; van der Kraan et al 1992). Further, TGF-β1 and 3 promote chondrogenesis of MSCs (Grimaud et al 2002; Li et al 2005; Schulz et al 2008; Puetzer et al 2010; Xu et al 2008).…”

“…[11][12][13][14] Members of the TGF-b superfamily play a major role in cartilage development and repair. [15][16][17] Last but not least, insulinlike growth factor 1 can stimulate the anabolic activity of chondrocytes and can induce chondrogenesis of mesenchymal stem cells. [18][19][20][21][22] In dentistry, PRGF-Endoret is already very much used (whether for uses for implantology, periodontal, or oral surgery) because it guarantees a better clinical recovery not only with a reduction of the postsurgery swelling and pain but also with a better osseointegration of the fixtures or in bone regeneration procedures.…”

“…Insulin-like growth factor-I (IGF-I), transforming growth factor b (TGFb)-1, TGFb-2, bone morphogenetic protein (BMP)-2, and BMP-7 have all been shown to improve chondrogenesis during 3D culture either in serum-free medium or in medium supplemented with fetal bovine serum (FBS). [6][7][8] We recently reported that culture of human nasal septal chondrocytes with pooled human serum (HS) results in increased proliferation and chondrogenesis compared with FBS. 9 The goal of this study was to identify growth factor combinations that improve chondrogenesis during 3D culture of human nasal septal chondrocytes in the medium supplemented with HS.…”
