Effect of growth factors on the activation of human Tenon’s capsule fibroblasts

Abstract: The present investigation strongly supports the hypothesis that PDGF-isoforms are major stimulators of proliferation of Tenon's capsule fibroblasts after glaucoma filtering surgery while TGF-beta-isoforms are essential for the transformation of Tenon's capsule fibroblasts into myofibroblasts.

“…Tenon’s capsule fibroblasts exposed to MMC have been reported to exhibit a significant decrease in the number of FGF-β receptors 4044. In our study, MMC and infliximab equally decreased the immunostaining intensity of FGF-β in the bleb region.…”

“…On the other hand, elevated levels of TGF-β are associated with increased fibrosis in several organs, including the heart, liver, kidney, lung, skin, and bone marrow 3942. TGF-β is considered to inhibit the growth of leukocytes and to stimulate the production of fibroblasts 43,44. TGF-β enhances resistance of the wound site to tension in corneal incisions in rabbits by enhancing the number of fibroblasts through chemotaxis, reducing the synthesis of matrix metalloprotease, and enhancing the synthesis of metalloprotease tissue inhibitor 42.…”

“…Degree and kinetics of proliferation differed, however, between media conditioned by the three cancer cell lines which is in line with the known fact that induction of fibroblastic stroma can differ between different tumours. We also identified in prostate cancer cell conditioned media growth factors (such as bFGF and PDGF) to which induction of fibroblast proliferation can be attributed (61,62). Basic fibroblast growth factor is already known to induce stromal growth within the human prostate (63).…”

“…Several studies reported that these factors are potent stimulators of HTF proliferation in vitro (Cunliffe et al, 1995;Denk et al, 2003;Knorr et al, 1997). Furthermore, inhibition of some of these molecules diminished growth of cultured human TF (Salas-Prato et al, 1996) and improved glaucoma filtration surgery outcome in an experimental glaucoma model (Turgut et al, 2014).…”
