Effect of genotype and medium on wheat (Triticum aestivum L.) anther culture

Abstract: Four winter wheat (Triticum aestivum L.) and two spring wheat cultivars were evaluated in anther culture on three to four different media for their ability to initiate callus and green plants. Five media were used in the experiment: stored-potato medium with Ficoll 400, fresh-potato medium with Ficoll 400, fresh-t ~tato medium with agar, fresh-potato liquid medium without agar or Ficol1400, and a one tep 85D12-3 medium. Greatly different frequencies of calli and/or green plants were obtained from different cul… Show more

“…Differences were observed in terms of shoot growth and production (Fazekas et al 1986;Juncker & Favre 1989), callus growth (Mikami & Kinoshita 1988;Myers et al 1989, Han-min et al 1990) and frequency of somatic embryogenesis (Chen et al 1987, Parrott et al 1989. Different genotypes of the same species respond differently to various culture media components such as coconut milk (Mathias & Simpson 1986), vitamins (Juncker & Favre 1989) and cytokinin concentration (Simpson & Bell 1989;Arnison et al 1990) and culture conditions such as cold pre-treatment (Powell 1988) and initial explant used (Chen et al 1987).…”

“…However, there remain problems ranging from low frequency of callus induction to large genotypic and environmental effects. The response of anthers cultured on medium is dependent on the genotype of the donor plant [7,9,10,12], and the environmental conditions under which these plants are grown [4,6,9]. Here we report on the genotypic and environmental effects on anther culture of various cultivars and lines of common wheat, most previously unscreened.…”
