IntroductionAgrobacterium tumefaciens is the most efficient way of introducing foreign genes into plants. Plants such as tobacco, potato, eggplant and other species of the family Solanaceae can easily be transformed by A. tumefaciens. However, cereals, legumes, and trees are recalcitrant to A. tumefaciens-mediated genetic transformation. Therefore, several strategies have been developed to increase genetic transformation frequency in recalcitrant plant species. These include the use of different bacterial density, inoculation and cocultivation methods, and explants (Kumar et al., 2002;Park, 2006;Sharma et al., 2009). In addition, the application of phenolic compounds to inoculation and cocultivation media also enhances transformation, even in recalcitrant plant species (Dutt and Grosser, 2009;Rashid et al., 2010).Squirting cucumber (Ecballium elaterium (L.) A.Rich.) is a poisonous plant that is used for medicinal purposes (Sezik, 1997). It contains different compounds, such as α-elaterin (cucurbitacin E), β-elaterin (cucurbitacin B), elatericine A (cucurbitacin D), and elatericine B (cucurbitacin I) in different plant organs. It also contains sterols, phenolic compounds, vitamins, flavonoids, alkaloids, resin, starch, amino acids, and fatty acids (Koç, 2002;Memişoğlu and Toker, 2002). Compounds found in leaves, stems, roots, and testa are poisonous and show antibacterial activities (Oskay and Sarı, 2007;Koca et al., 2010;Adwan et al., 2011). Similarly, Adwan et al. (2011) also reported antibacterial activity of ethanol extracts obtained from mature dried fruit of squirting cucumber, which exerted negative effects on the growth of Staphylococcus aureus and Candida albicans. However, in our unpublished preliminary studies, we found that the juice of mature fruits induced growth of E. coli and Staphylococcus aureus. Therefore, the present study was designed, for the first time, to check the effects of fresh juice of mature fruits of squirting cucumber on growth of A. tumefaciens and its gene transfer efficiency to tobacco and potato plants.

Zostera marina L. is one of marine flowering plants, which plays an important role in coastal ecosystems by generating oxygen and organic substance and providing a habitat for other organisms. However, its residues on seashore have caused serious environmental problems in some coasts worldwide. In this paper, crude extract of Z. marina L. was prepared using methanol-water (1:1, v/v) and further separated into petroleum ether fraction, ethyl acetate fraction, n-butanol fraction and water fraction. The antibacterial activity and antioxidant activity of different fractions from Z. marina L. were evaluated in order to provide information for utilizing the marine plant. Antibacterial bioassay showed that n-butanol fraction was effective only for Staphylocccus aureus, petroleum ether fraction was effective for S. aureus and Bacillus anthracis, ethyl acetate fraction was effective for S. aureus, B. anthracis, Diphtheroid bacilli and Staphylococcus epidermidis, while water fraction had no effect on all the tested strains. Antioxidant assay indicated that all of the four fractions showed α-diphenyl-β-picrylhydrazyl (DPPH) radical-scavenging activity and Fe 3+ reducing activity, and ethyl acetate fraction exhibited the maximum activity.

Intra-peritoneal administration of Ecballium elaterium diminishes postoperative adhesions

PURPOSE:To evaluate the effects of Ecballium elaterium (EE), Elaterium officinarum, in postoperative intraperitoneal adhesions in rats. METHODS:Thirty rats were divided into three groups and underwent midline laparotomy under 35 mg/kg ketamine and 5 mg/kg xylazine anaesthesia. In group 1 (n=10), the sham operation group, the abdominal walls were closed without any process. In group 2 (n=10), the control group, the antimesenteric border of the ceacum and the corresponding parietal peritoneum were abraded with dry sterile gauze. In group 3 (n=10), the EE group, 2.5 mg/kg dose of EE was administered as intraperitoneally to the rats after abrasion. All rats were sacrificed on postoperative day 15. Samples were obtained RESULTS:The adhesion score was significantly decreased in the EE group (p=0.001) in comparison with the control group.Microscopically, the EE and sham groups were significantly lower than that of the control group (p<0.001 and p=0.000, respectively).Furthermore, the measurement of tissue levels of hydroxyproline was significantly lower in the sham and EE groups compared to the control group (sham group: 47.6 ± 10.6, EE group: 62.9 ± 9.7, Control group: 84.2 ± 22.1 mg /L/g-tissue). CONCLUSION:The grade and severity of abdominal adhesion could be significantly reduced through administered Ecballium elaterium and therefore be a suitable anti-inflammatory agent for the prevention of postoperative peritoneal adhesion in the future.
