<span style="font-family: arial,helvetica;">Foot-and-mouth disease (FMD) is an economically important disease of cloven-hoofed animals that is primarily controlled by vaccination of susceptible animals and movement restrictions for animals and animal-derived products in South Africa. Vaccination using aluminium hydroxide gel-saponin (AS) adjuvanted vaccines containing the South African Territories (SAT) serotypes has been shown to be effective both in ensuring that disease does not spread from the endemic to the free zone and in controlling outbreaks in the free zone. Various vaccine formulations containing antigens derived from the SAT serotypes were tested in cattle that were challenged 1 year later. Both the AS and ISA 206B vaccines adjuvanted with saponin protected cattle against virulent virus challenge. The oilbased ISA 206B-adjuvanted vaccine with and without stimulators was evaluated in a field trial and both elicited antibody responses that lasted for 1 year. Furthermore, the ISA 206 adjuvanted FMD vaccine protected groups of cattle against homologous virus challenge at very low payloads, while pigs vaccinated with an emergency ISA 206B-based FMD vaccine containing the SAT 1 vaccine strains were protected against the heterologous SAT 1 outbreak strain.</span>

Liquid array technology has previously been used to show proof of principle of a multiplexed nonstructural protein serological assay to differentiate foot-and-mouth disease virus-infected and vaccinated animals. The current multiplexed assay consists of synthetically produced peptide signatures 3A, 3B, and 3D and the recombinant protein signature 3ABC in combination with four controls. To determine the diagnostic specificity of each signature in the multiplex, the assay was evaluated against a naive population (n ‫؍‬ 104) and a vaccinated population (n ‫؍‬ 94). Subsequently, the multiplexed assay was assessed by using a panel of bovine sera generated by the World Reference Laboratory for foot-and-mouth disease in Pirbright, United Kingdom. This serum panel has been used to assess the performance of other singleplex enzyme-linked immunosorbent assay (ELISA)-based nonstructural protein antibody assays. The 3ABC signature in the multiplexed assay showed performance comparable to that of a commercially available nonstructural protein 3ABC ELISA (Cedi test), and additional information pertaining to the relative diagnostic sensitivity of each signature in the multiplex was acquired in one experiment. The encouraging results of the evaluation of the multiplexed assay against a panel of diagnostically relevant samples promote further assay development and optimization to generate an assay for routine use in foot-and-mouth disease serological surveillance.Liquid array technology allows simultaneous measurement of the relative responses of multiple signatures to a challenge sample (19). This technology has proven successful for multiple applications; antigen and nucleic acid-based biological threat agent detection (27, 28) and serological assays (3,16,20,23,35,42,43) are some examples. The use of such multiplexing technology has time, cost, and manpower benefits over multiple singleplex analyses, in addition to increased confidence in the results obtained. Multiple-signature evaluation provides more confidence when obtaining a conclusive result; it eliminates variations that may occur when using a series of singleplex assays to obtain a comparative result, and it allows controls in every sample. The liquid array consists of beads that are embedded with precise ratios of red and infrared fluorescent dyes yielding 100 bead sets, each with a unique spectral address. Analyte that is captured on a modified bead is detected by a detector reagent indirectly labeled with a fluorescent reporter. Each optically encoded and fluorescently labeled bead is then tested with a flow cytometer. A classification laser (635 nm) excites the dye molecules inside the bead and classifies the bead into its unique bead set. A reporter laser (532 nm) excites the bound fluorescent reporter and quantifies the assay at the bead surface. Foot-and-mouth disease (FMD) is a highly contagious viral disease affecting biungulate species. Commercial vaccines, formulated with inactivated FMD virus (FMDV), are regularly used worldwide to control the disease. Here, we studied the generation of antibody responses in local lymphoid tissues along the respiratory system in vaccinated and further aerosol-infected cattle. Animals immunized with a high-payload monovalent FMD vaccine developed high titers of neutralizing antibodies at 7 days postvaccination (dpv), reaching a plateau at 29 dpv. FMDV-specific antibody-secreting cells (ASC), predominantly IgM, were evident at 7 dpv in the prescapular lymph node (LN) draining the vaccination site and in distal LN draining the respiratory mucosa, although in lower numbers. At 29 dpv, a significant switch to IgG1 was clear in prescapular LN, while FMDV-specific ASC were detected in all lymphoid tissues draining the respiratory tract, mostly as IgM-secreting cells. None of the animals (n ‫؍‬ 10) exhibited FMD symptoms after oronasal challenge at 30 dpv. Three days postinfection, a large increase in ASC numbers and rapid isotype switches to IgG1 were observed, particularly in LN-draining virus replication sites already described. These results indicate for the first time that systemic FMD vaccination in cattle effectively promotes the presence of anti-FMDV ASC in lymphoid tissues associated with the respiratory system. Oronasal infection triggered an immune reaction compatible with a local anamnestic response upon contact with the replicating FMDV, suggesting that FMD vaccination induces the circulation of virusspecific B lymphocytes, including memory B cells that differentiate into ASC soon after contact with the infective virus. F oot-and-mouth disease (FMD) is a highly contagious and acute viral disease affecting a wide range of economically important livestock species (1). All domestic biungulates are susceptible to infection with the FMD virus (FMDV); in addition, a number of wildlife species may act as reservoirs for the virus under particular ecological conditions (2). Lethality has been described for young animals and certain FMDV strains (3).
