Effect of Detergents Based on Sodium Dodecyl Sulfate on Functional Metrics of Frozen–Thawed Collared Peccary (Pecari tajacu) Semen

Abstract: We evaluated the effects of detergents based on sodium dodecyl sulfoxide (SDS) on the functional parameters of collared peccary frozen–thawed sperm. Semen aliquots from ten individuals were diluted in a Tris–egg yolk–glycerol extender alone or with 0.5% Equex STM® paste or SDS (at 0.1%, 0.3% or 0.5% (v/v) concentration). Samples were fast frozen in liquid nitrogen with a post-thaw evaluation of motility, membrane functionality and integrity, mitochondrial activity, sperm binding ability and thermal resistance.… Show more

“…In general, the data obtained in the present work are similar to the results obtained previously in the freezing of collared peccary semen, also without the use of antioxidants in the diluent (Moreira et al., 2022, 2023). Regarding ROS analysis, however, only recently another study presented the evaluation of this parameter for fresh semen using the same methodology described here (Santos et al., 2023).…”

“…In this sense, different efforts were conducted to better understand the physiological aspects of collared peccary semen (Peixoto et al., 2012), and to improve cryopreservation protocols that enable the formation of a biobank (Moreira et al., 2023). Bearing in mind that the fluid from the accessory glands of the genital tract can play an important role in semen freezeability (Moreira et al., 2022), different aspects of collared peccary seminal plasma were deciphered, such as the identification of small amounts of superoxide dismutase (SOD), while catalase (CAT) was not found in such fluid (Santos et al., 2018).…”

“…For the cryopreservation protocol, the cooling samples already diluted in TRIS + yolk (20%) and PRIMXcell Ultra were removed from the respective cooling systems at specific intervals (4, 24, and 48 h), and 6% glycerol was added (also at 5 °C), resulting in a final concentration of 3% glycerol in the extender [ 4 ]. The samples were then filled into 0.25 mL straws, which were then placed on a ramp (height 5 cm), exposed to liquid nitrogen for 5 min, and stored in a cryobiological cylinder at −196 °C [ 1 ]. After a week, the samples were thawed in a water bath at 37 °C [ 4 ], and sperm parameters were assessed.…”

“…Given the persistence of threats to the survival of various free-living species, there is a growing need for efficient strategies to conserve genetic material [ 1 ]. The creation and establishment of biological sample repositories (biobanks) are emerging as promising options for the preservation of wild animals, especially for species threatened with extinction [ 2 ].…”
