Effect of dehydroepiandrosterone sulfate on interleukin-8 receptor during cervical ripening

Abstract: We investigated the effects of dehydroepiandrosterone sulfate (DHA-S) on the production of interleukin-8 (IL-8) and expression of the interleukin-8 receptor (IL-8 R) in human cervical tissue. DHA-S increased the levels of IL-8 in cultured human cervical fibroblasts and in the supernatant in a timeand dose-dependent manner. DHA-S induced IL-8 and IL-8 R expression in human cervical fibroblasts and human pregnant cervical tissue at term in a dose-dependent manner. In addition, it induced the expression of IL-8 R… Show more

“…Numerous human and animal studies have explored the role of androgens in the cervix, particularly during pregnancy (Mochizuki et al , 1978a, b; Sasaki et al , 1982; Mochizuki and Maruo, 1985; Takahashi et al , 1984; Sakyo et al , 1986, 1987; Yamashita et al , 1991; El Maradny et al , 1996; Kanayama et al , 1998; Ji et al , 2008). These studies formed the basis of a hypothesis, whereby androgens regulate cervical remodelling, particularly CR at term.…”

“…Indeed, DHEAS action on CR has been correlated with increased levels of IL-8 cytokine involved in chemotaxis of neutrophils recruited in cervix towards term (Kanayama et al , 1998; Maymon et al , 2000). …”

“…This shows that DHEA-S might have a synergistic effect on IL-8 action. Additionally, it was shown that DHEA-S can stimulate the expression of IL-8 and IL-8 receptors and enhance HA production in the cervical stroma [ 107 , 108 ]. As mentioned before, HA is a strong IL-8 synthesis stimulator.…”

“…Among steroid hormones that influence remodeling of uterine cervix, dehydroepiandrosterone sulphate (DHEA‐S), and dihydrotestosterone (DHT) stand out 7 . DHEA‐S has role in activation of neutrophils and macrophages, that is, it stimulates activation of the proteolytic enzymes in uterine cervix 8–10 . Progesterone (P4), DHT, dehydroepiandrosterone (DHEA), and DHEA‐S on the other side, make impact on the uterine contractility 11 …”
