Effect of dam methylation on the activity of the E. coli replication origin, oriC.

Abstract: Methylation of GATC sites by the dam methylase is required for efficient initiation of DNA replication at the replication origin, oriC, of Escherichia coli. This is demonstrated by the inability of minichromosomes to be maintained in dam mutant strains. The requirement for methylated GATC sites is less stringent in vitro than in vivo. The time required for complete methylation of the origin region apparently determines the minimal spacing of replication forks on the chromosome.

“…dam methylase selectively methylates the adenine residues of the sequence GATC of newly synthesized DNA [3]. We chose dam methylnse for the following reasons: (4 it is a small monomeric enzyme of 30 kDa ]4]; (ii) it needs only ~~~~~~~osylrnethionine for its action [4]; (iii) it has several significant biological roles: &sn rhethylase discriminate the parent from the dawhter stt%nd of DNA dnring ~is~at~~ repair f5& is ~~~~~~~d in replication [6] …”

“…Previous experiments demonstrated that dam bacterial strains are poorly transformed by fully methylated and hemimethylated plasmids dependent on either the chromosomal replication origin oriC or the pMBl plasmid origin (28,38,39). This is due to the lack of replication of hemimethylated molecules, which accumulate in dam strains transformed by methylated plasmids (38).…”

“…Methylation at ori-C also influences the local DNA structure and stability (62, 66 -71) and its interaction with replication proteins (72). Hemimethylated ori-C is refractory to further initiation events (73)(74)(75) due to the inability of DnaA protein to access ori-C (76), thus preventing premature reinitiation when free DnaA levels are still high (64,65,76). OriC persists in this hemimethylated/sequestered state for ϳ8 min, or about 10 times longer than for most other sites on the E. coli chromosome (64,65).…”
