Effect of Cyclophosphamide and Its Combination with Metformin on the Survival Rate in Mice

Abstract: Background: Cyclophosphamide (CYP), an alkylating chemotherapeutic agent, is widely used to treat several types of cancer. Its toxic effects are well-established and include hepatotoxicity, nephrotoxicity, and bone marrow suppression. Metformin (MET) is an anti-diabetic medication that is considered a first-line therapy for type 2 diabetes mellitus. In this study, we aimed to investigate the effect of co-administration of MET on CYP-induced toxicity by recording the survival rate in mice. Methods: Fifty mice (… Show more

“…Примечательно, что результаты настоящего исследования согласуются с экспериментальными данными о снижении выживаемости мышей при сочетанном применении циклофосфамида и метформина [24], а также данными, демонстрирующими снижение под влиянием метформина резистентности опухолей к циклофосфамиду [25], паклитакселу и цисплатину [26].…”

“…In addition, the idea of using it in other age-related diseases has been suggested [ 7 ]. Regarding the mechanism of action of metformin, there is much controversy, but the activation of AMPK is the result of the interaction of the biguanide compound with the mitochondrial complex I [ 8 , 9 , 10 ]. However, metformin is also associated with the modulation of other cellular signaling pathways, including caspase 3, Bcl-2, and mTOR pathways, which may explain the anti-apoptotic, antioxidant, and pro-autophagic effects [ 11 ].…”

“…In spite of its beneficial effects, it has many side effects (Farkas et al, 2019) even when given at low doses which include petechial hemorrhage in the small bowel, abdominal pain, diarrhea, and inflammation (Alhowail et al, 2019). The CP caused severe tissue oxidative stress and massive cellular damage, consequently increasing apoptosis and death of both cancer and healthy cells (Abdel-Hafez et al, 2017b).…”
