Effect of culture time on secondary metabolite production from MeJA-stimulated in vitro roots of Hyoscyamus niger L.

Abstract: This study was conducted to determine the effects of different culture times on root growth and accumulation of secondary metabolites in adventitious roots from Hyoscyamus niger petioles. For this purpose, adventitious roots transferred to culture media containing 1 mM methyl jasmonate (MeJA) were harvested at 11 different culture times, starting from day 1 to day 30. After harvest, the fresh and dry weights of the roots and the root growth index as parameters of root growth, as well as the amounts of tropane … Show more

“…According to the culture method, Wang et al (2022) recently observed that the MeJA-treated Chinese chives had substantial variations in primary and secondary metabolites. Similar outcomes were also reported by other researchers, who claimed that using MeJA as an elicitor in the culture increased the synthesis of the secondary metabolites (Rodrıǵuez-Sańchez et al, 2020;Deveci et al, 2022;Wang et al, 2022).…”

“…Despite the therapeutic effects, these compounds also have toxic effects on humans. Scopolamine and hyoscyamine can cause intoxication by paralyzing the nerve endings of the parasympathetic system (Deveci et al 2022).…”

“…The dependence of tropane alkaloid and phenol accumulation on the duration of cultivation and L-phenylalanine treatment in the in vitro AR culture of Hyoscyamus niger L. was shown [52]. The effect of the culture period on the yield of secondary metabolites with an optimal 15-day period is shown in the in vitro AR of Hyoscyamus niger L. [53]. The HPLC of the in vitro culture of Lang Bian ginseng showed that only twenty-week-old in vitro rhizomes had ginsenoside Rg1, Rd and Rb1 [54].…”
