Effect of Cryopreservation Method Supported with Biochemical Analyses in the Axillary Bud of Jewel Orchid, Ludisia discolor

Abstract: This study investigated conserving an endangered terrestrial jewel orchid Ludisia discolor, using in vitro grown axillary buds. Excised segments of axillary buds (4–5 mm in length) were precultured on a modified Murashige and Skoog (MS) medium supplemented with 0.2 M sucrose for 24 h and osmoprotected in a loading solution for 20 min. Then, axillary buds were dehydrated in plant vitrification solution 2 (PVS2) for 10 min at 0 °C and incubated in liquid nitrogen for 1 h. Subsequently, axillary buds were rewarme… Show more

“…An important aspect of understanding the stress associated with cryopreservation may be achieved through biochemical studies of oxidative stress (Prudente & Paiva, 2017). Even if there are many studies concerning the antioxidant activities during cryopreservation (Rahmah et al, 2015;Liu et al, 2021;Burkhan et al, 2022), none of them referred to sterile airflow dehydration in gametophyte ferns. For this purpose, the activity of catalase and superoxide dismutase of cryostorage gametophytes exposed airflow dehydration and were determined.…”

“…In our work, 0.3 M sucrose is the best concentration for the preculture of P. gigantea seeds. But for other species such Jewel Orchid, Ludisia discolor sucrose at 0.2 M for 24 h was the best sucrose preculture treatment that enhanced germination (Burkhan et al, 2022). While Paphiopedilum insigne (Wall.…”

“…The increasing demand and inadequate management practices have led to its scarcity due to overharvesting (Poobathy et al 2019). Consequently, wild L. discolor is dying out due to overexploitation in recent years and has been reported in several countries, such as China (Su et al 2017), Malaysia (Burkhan et al 2022), and Vietnam (Nguyen and Phung 2023). Moreover, the powder of L. discolor in the market was mixed with powders of other jewel orchid species, such as Anoectochilus roxburghii, A. formosanus, or Goodyera schlechtendaliana leading the reduced effectiveness of treatment efficacy (Chai et al 2022).…”
