Effect of corticosteroid treatment on interleukin‐1 and tumour necrosis factor secretion by monocytes from subjects with asthma

Abstract: Peripheral blood monocytes (PBM) may be activated in asthmatic patients, a condition usually reverted by corticosteroid (CS) treatment. In the present research we have evaluated the spontaneous or lipopolysaccharide (LPS)-induced production of interleukin 1 (IL-1) and tumour necrosis factor (TNF) by PBM obtained from 14 asthmatic subjects during an asthmatic attack and after 1 week of CS treatment. The control group included 20 healthy volunteers. PBM obtained during severe asthma showed a pattern of IL-1 and … Show more

“…First, the manifestations of asthma are, at least in part, channeled through the actions of IL-1␤ and TNF-␣ (11,12). Second, the efficacy of GCs in asthma is, at least in part, through its effect on the expression of genes that are modulated by proinflammatory cytokines (12,13). In this study, we show that GC responders can be separated from nonresponders with meaningful accuracy by using the expression levels of only a few genes.…”

“…TNF-u and IL-I~are key regulators of immune and inflammatory genes (24)(25). Conversely, IL-4 is a pivotal cytokine that is produced by Th2 cells and associated with allergic diseases that support Th2 development (26)(27).…”

“…Supernatants from PBM were obtained according to a previously described method [22]. Briefiy, PBM were seeded at a concentration of 2 x 10^/ml of cRPMI for 48 h without or with LPS (10/ig/mi) or Timothy grass pollen.…”
