Effect of Constitutive miR164 Expression on Plant Morphology and Fruit Development in Arabidopsis and Tomato

Abstract: Several miRNAs are conserved in different plant families, but their abundance and target genes vary between species, organs, and stages of development. Target genes of miRNAs are mostly transcription factors, involved in the control of many plant developmental processes, including fruit development. MiR164 is a conserved miRNA, highly expressed in fruits, and is validated to target a subset of genes of the NAC-domain transcription factor gene family. The objective of this work was to analyze the phenotypic eff… Show more

“…By comparing dormant stages and bud break stages, more than half of the microRNAs were positively correlated with their target mRNAs. Positive correlation of expression between microRNAs and their targets has also been observed in other plant species (Lopez-Gomollon et al, 2012;Chen et al, 2015;Cárdenas et al, 2017;Neller et al, 2018;Liang et al, 2019). As the target genes change in expression over developmental stages, additional regulatory factors including transcription factors in tandem with microRNAs are likely involved.…”

“…Nonetheless, the only sequence-based in silico homology approaches to identify potential miRNAs in new plant species may yield false-positive results, and hence consideration must be given to the secondary structures as well as other parameters of the pre-miRNAs such as length, GC content, Minimum Folding Free Energy (MFE), and Minimum Folding Free Energy Index (MFEI) in order to increase the precision of the computational prediction by discriminating from other coding or non-coding RNAs [6,12,13]. However, experimental confirmation of the predicted miRNAs is strongly recommended [6,14,15].…”

“…AAP4 was found to be upregulated in mango ginger rhizome when compared to ginger leaves and rhizome. Mtr-miR164 which is its miRNA is conserved, highly expressed in fruits, and is validated to target a subset of genes of the NAC-domain transcription factor gene family 56,57 . It was found to have signi cant differential expression with mango ginger when compared with ginger in both leaves and rhizome (Fig.…”
