Here, we investigate the apoptotic effect of allicin, the predominant component of freshly crushed garlic, on neuroblastoma cells. In this paper, the authors have first assessed the effect of allicin on human neuroblastoma SK-N-SH cells and then investigated the underlying mechanism. The results indicate that allicin suppresses SK-N-SH cell growth in a dose-dependent and time-dependent manner and that 5 μmol/l of allicin leads to a significant increase in apoptotic rate with annexin-V/PI double staining. Western blot analysis shows that treatment with allicin-induced apoptosis through activation of caspases-3 and 9. Phosphorylation of p38 MAPK contributes to allicin-induced apoptosis upstream of caspase activation. Using p38 MAPK inhibitor, the authors discovered that p38 MAPK activation subsequently induces the release of cytochrome-c from mitochondria into the cytosol. Taken together, the results demonstrate that allicin can activate the p38 MAPK pathway, which leads to mitochondrial release of cytochrome-c, thus inducing SK-N-SH cell apoptosis. Overall, this study suggests that allicin may be used as one of the novel pharmacological treatment strategies in neuroblastoma.

Background and Objectives: Chronic wounds represent a significant health problem. Novel therapeutic approaches to wound healing are required. Garlic (Allium sativum) was considered to exert antioxidant, antibacterial, and anticholesterol activities. Aim of the work: The present work aimed at investigating the possible therapeutic effect of garlic extract (GE) on induced wound in male albino rat. The possible role of endogenous stem cells was also determined. Materials and Methods: 26 adult male albino rats were divided into Group I (Control group): 6 rats not exposed to wound induction.Group II (Wounded group): 10 rats were exposed to wound induction and subdivided into subgroups IIa and IIb, 5 animals each, were sacrificed 2 and 3 weeks following wound induction. Group III (Wounded and GE treated group): 10 rats were exposed to wound induction and subdivided as in group II. 0.5 ml of 1% GE was locally applied following wound induction. Skin specimens (including the wound and a margin of normal skin around) were subjected to histological, immunohistochemical, morphometric and statistical studies. Results: Morphological changes, indicating inflammation and degeneration, were found in wounded rats. These changes regressed remarkably by the use of GE. The regression of apoptosis was confirmed by caspase3 immunostaining and the activation of stem cells was proved by CD44 immunostaining. Conclusions: GE therapeutic effect was proved to be related to both caspase3 pathway inactivation and MSC migration activation. GE was suggested to be applied in appropriate concentrations in the preparation of pharmaceutical products to be used safely on epithelial cells in wound care.

Background Side population (SP) cells, which have similar features to those of cancer stem cells, show resistance to dexamethasone (Dex) treatment. Thus, new drugs that can be used in combination with Dex to reduce the population of SP cells in multiple myeloma (MM) are required. Diallyl thiosulfinate (DATS, allicin), a natural organosulfur compound derived from garlic, has been shown to inhibit the proliferation of SP cells in MM cell lines. Therefore, we investigated the effect of a combination of DATS and Dex (DAT + Dex) on MM SP cells. Methods SP cells were sorted from MM RPMI-8226 and NCI-H929 cell lines using Hoechst 33342-labeled fluorescence-activated cell sorting. The growth of SP cells was evaluated using the cell counting kit-8 assay. Cell cycle and apoptosis assays were conducted using a BD Calibur flow cytometer. miRNA expression was measured using quantitative reverse transcription-polymerase chain reaction. Phosphoinositide 3-kinase (PI3K), phosphorylated AKT (p-AKT), AKT, p-mechanistic target of rapamycin (mTOR), and mTOR levels were measured using western blot analysis. Results Our results showed that the combination of DATS+Dex inhibited sphere formation, colony formation, and proliferation of MM SP cells by inducing apoptosis and cell cycle arrest in the G1/S phase. In addition, the combination of DATS+Dex promoted miR-127-3p expression and inhibited PI3K, p-AKT, and p-mTOR expression in SP cells. Knockdown of miR-127-3p expression weakened the effect of DATS+Dex on cell proliferation, colony formation, apoptosis, and cell cycle of MM SP cells. Additionally, knockdown of miR-127-3p activated the PI3K/AKT/mTOR signaling pathway in MM SP cells cotreated with DATS+Dex. Conclusion We demonstrated that cotreatment with DATS+Dex reduced cell proliferation, promoted apoptosis, and caused cell cycle arrest of MM SP cells by promoting miR-127-3p expression and deactivating the PI3K/AKT/mTOR signaling pathway.
