Effect of cleistanthin A and B on adrenergic and cholinergic receptors

Abstract: Objective:The aim was to study the in vitro and in silico interactions of cleistanthin A and B on the adrenergic and cholinergic receptors using isolated animal tissues and bioinformatics tools.Materials and Methods:The alpha adrenergic receptor activities of cleistanthin A and B were studied in vitro using a guinea pig vas deferens preparation. The beta adrenergic receptor activities of cleistanthin A and B on an isolated rat heart were studied in vitro using a modified Langendorff apparatus. The effects of c… Show more

“…It was reported in a study conducted on isolated rat heart that both cleistanthins A and B had no effect on beta 1-adrenoceptors. 7 This suggests that the blockade of phenylephrine induced relaxation could be due to alpha 1-adrenoceptor antagonism. From the present study, it is evident that cleistanthins A and B effectively blocked the phenylephrine induced relaxation and this effect could be attributed to blockade of alpha 1-adrenoceptors.…”

“…This finding is in agreement with the effect of cleistanthins A and B on rat vas deferens where both the compounds prevented the phenylephrine induced contractions. 7 We have collected all the blood samples under general anaesthesia and any undue stress induced endogenous epinephrine release was avoided. We have estimated the plasma insulin levels using an ultra-sensitive rat ELISA kit that can detect insulin levels up to ng/ml precise when compared to conventional radio-immuno assay technique.…”

“…[ 23 ] Parasuraman and Raveendran reported that cleistanthins A and B had an alpha-adrenergic receptor blocking property in isolated guinea pig vas deferens. [ 24 ] It was suggested that cleistanthins A and B reduce BP, and it could be due to their alpha-adrenergic receptor blocking property. [ 22 ] The effect of cleistanthins, A and B on BP in repeated doses, was not studied, and their antihypertensive effect was not evaluated using animal models of hypertension.…”

“…Cleistanthus has been shown to contain bioactive lignan compounds such as cleistanthins A and B (from Cleistanthus collinus); these were found to be cytotoxic against several tumor cell lines [7,8]. Also, cleistanthins A and B inhibited the actions of the alpha adrenergic receptor and the nicotinic cholinergic receptor [9]. In Vietnam, 14 species of Cleistanthus have been identified and, importantly, some of them were used in traditional medicine [10].…”
