Effect of cetirizine in a conjunctival provocation test with allergens

Abstract: Cetirizine is a new selective H1-antagonist. In a double-blind and placebo-controlled study, 11 subjects underwent a conjunctival provocation test (CPT) with a grass pollen extract to determine the effect of cetirizine on allergen-induced itching and redness of the eye. Results of CPTs were reproducible, as no difference was observed between the CPTs performed without any pre-treatment and the CPTs after placebo. Compared to placebo, cetirizine administered orally (10 mg twice a day for 4 days) and in double-b… Show more

“…747 Oral antihistamines, intranasal antihistamines, oral anti-LT agents, intranasal corticosteroids, and allergen immunotherapy are treatments for allergic rhinitis that have been reported to relieve associated ocular allergy symptoms in controlled trials. [37][38][39][40][41][42][43][46][47][48][49][50][51][52][53][54] In systematic reviews of randomized controlled studies, intranasal corticosteroids compared with oral antihistamines 53,54,748 and intranasal corticosteroids compared with intranasal antihistamines 46 were not significantly different in relieving eye symptoms. In placebo-controlled studies of adults, fluticasone furoate nasal spray has been demonstrated to reduce significantly ocular symptoms associated with seasonal allergic rhinitis.…”

“…In skin chamber studies, cetirizine was found to inhibit eosinophil migration [2] and histamine release [3] following the injection of pollen in sensitized subjects. In clinical studies cetirizine reduced the oceular reaction to instilled allergen [4] and in pollen sensitive asthmatic subjects it prevents the deterioration in asthma symptoms observed during the pollen season [5]. It has been suggested that inhalation of cetirizine may be more effective than oral therapy in asthmatic subjects since inhaled cetirizine has been shown to inhibit exercise induced bronchoconstriction while oral therapy had no effect [6].…”

“…It has been postulated that the mechanism(s) of action of drugs used to treat allergic diseases are likely to be elu- $36 cidated by studying the events following allergen-specific CPT [2][3][4]7].
