Effect of Bradykinin Receptor Antagonists on Vincristine- and Streptozotocin-Induced Hyperalgesia in a Rat Model of Chemotherapy-Induced and Diabetic Neuropathy

Abstract: The role of bradykinin receptor blockade in the development of neuropathies caused by diabetes mellitus and vincristine was examined. The effects of a potent and selective B1 receptor antagonist (des-Arg10-HOE 140) as well as a specific antagonist of B2 receptors (HOE 140) were investigated. Both agents significantly decreased hyperalgesia caused otherwise by vincristine. In a diabetic neuropathy model, both agents almost completely suppressed hyperalgesia in the first 10 days … Show more

“…Acetyl-L-carnitine can prevent mPTP opening (Pastorino et al, 1993) and is associated with a reduction in paclitaxel, oxaliplatin and bortezomib-induced CIPN when administered prophylactically in rodents (Jin et al, 2008; Bujalska and Makulska-Nowak, 2009; Carozzi et al, 2010b; Xiao et al, 2012; Zheng et al, 2012). …”

“…79 More interestingly, simultaneous administration of indomethethacin or celecoxib with selective bradykinin B1or B2 antagonists produces an additive anti-hyperalgesic effect on the vincristine-induced neuropathic pain model, 80 whilst celecoxib alone had no effect on alleviating this type of pain. 57 These results indicate that the interaction between the activation of bradykinin receptors and cox pathways plays an important role in the development of chemotherapy-induced peripheral neuropathic pain.…”

“…Pre-emptive analgesia with with lower dose of indomethacin and celecoxib significantly suppressed vincristin-induced hyperalgesia. 80-82 Therefore, the pre-emptive strategy may play an important role in prevention of peripheral neuropathic pain induced by chemotherapeutic agents.…”
