Effect of benzo[a]pyrene on the expression of miR‐126, miR‐190a and their target genes EGFL7, TP53INP1 and PHLPP1 in primary endometrial cells

Abstract: The main topic of this study was to investigate the effect of benzo[a]pyrene (BP) on microRNAs and their target genes expression levels in primary cell cultures from normal and malignant endometrial tissue. MicroRNA-126 (miR-126) and miR-190a were most sensitive to BP treatment. The treatment of both cultures with BP was accompanied by a decrease of miR-126 level and an increase of EGFL7 gene expression level. BP-induced upregulation of miR-190a was detected only in normal cells and it was accompanied with dec… Show more

“…Mechanism study have shown that miR-145 functioned as EMT-suppressor in NSCLC by targeting MTDH, Yin Q also found that the over-expression of mir-145 could inhibit TGF- β-induced EMT in non-small cell lung cancer cells ( 60 ). MiR-126-3p located in the host gene of epidermal growth factor domain 7 (EGFL7) of human chromosome 9q34.3, was also involved in different rearrangements in various cancers ( 61 ). One of the most notable targets in lung cancer is EGFL7 ( 62 , 63 ).…”

“…Interestingly, benzo[a]pyrene promotes the proliferation of normal cells and exacerbates apoptosis signaling in cancer cells. BP-induced upregulation of miR-190-5p was detected in normal cells accompanied with downregulation of mRNA levels of TP53INP1 and PHLPP1 genes, which demonstrates that miR‐190-5p is possibly involved in the general cellular response to benzo[a]pyrene [40]. These findings show that the relationship between miR-190-5p and cell proliferation is complex and that miR-190-5p might have different effects in various types of human cancers, which requires further investigation to address these discrepancies.…”

“…For example, Kim showed that 1 μM Bap had no cytotoxic effect on endometrial carcinoma cells, while Chanyshev demonstrated that Bap evoked a significant level of endometrial carcinoma cells death (40%) after 24 h-treatment at the same concentration. 11,12 These experiments were conducted above the aqueous solubility limit of Bap. 13 We speculated that Bap existed in the form of nanoclusters and the misunderstanding of Bap's form was probably responsible for the inconsistent results of the previous studies.…”
