Effect of Basal Medium, Explants Size and Density on the In Vitro Proliferation and Growth of Date Palm (Phoenix dactylifera L.) Cultivar ‘16-bis’

Abstract: The effect of basal medium, explant size and density on shoot multiplication, growth, rooting and acclimatization of date palm cv. '16-bis' was evaluated. Bud clusters of different sizes (2, 3, 4 and 5 buds per cluster) were cultured at density of 1, 2, 3 and 4 clusters on Murashige and Skoog medium (MS), woody plant medium (WPM) and Nitsch medium (NM) supplemented with 0.5 mg/L 2-naphthoxyacetic acid and 0.5 mg/L kinetin for three months (multiplication phase). Separated shoots of different sizes (<3 cm; 3 to… Show more

“…In cv. 16-bis, MS medium showed higher multiplication efficiency than woody plant medium and Nitsch medium (Mazri 2013 ). In the present work, the combination of 825 mg/L NH 4 NO 3 , 1900 mg/L KNO 3 , 220 mg/L CaCl 2 ·2H 2 O, 170 mg/L KH 2 PO 4 and 370 mg/L MgSO 4 ·7H 2 O considerably stimulated the formation of adventitious buds.…”

“…However, the majority of works focused on the effects of plant growth regulators (PGRs). The effects of other factors such as mineral salts, l -glutamine, myo-inositol and carbon source have been scarcely evaluated for date palm organogenesis (Al-Khateeb 2008a ; Mazri 2013 , 2014 ; Mazri and Meziani 2013 ).…”

“…The highest multiplication ratio occurred when the plant density was lowest (6 buds per vessel). In sugarcane [ 26 ], hosta [ 27 ], garlic [ 28 ], and palm culture [ 29 ], the multiplication increased with lowered plant density. Increasing P from 1.25 mM, which is the concentration of P in the standard MS medium and the lowest concentration tested to 6.25 mM P produced more plants.…”

“…Moreover, explants between one and three cm prepared from shoot tips of species from Cryptomeria, Sequoia and Chamaecyparis genera of the Cupressaceae family gave better results compared to smaller explants (Isikawa, 1987;Ball, 1987;Kurz, 1989). In general, the effect of explant size on regeneration in plants is well documented (Mazri, 2013;Rehman, 2014;Plaza-Wüthrich et al, 2015). In the present study, the main effects of the three media averaged across explant size, indicated a contrasting response of the number of axillary shoots formed and mean shoot length.…”
