Effect of ATRA and ATO on the expression of tissue factor in NB4 acute promyelocytic leukemia cells and regulatory function of the inflammatory cytokines TNF and IL-1β

Abstract: The characteristic hemorrhages of acute promyelocytic leukemia (APL) are caused in part by the high expression of tissue factor (TF) on leukemic cells, which also produce TNF and IL-1β, proinflammatory cytokines known to increase TF in various cell types. Exposure of NB4 cells, an APL cell line, to all-trans retinoic acid (ATRA) or arsenic trioxide (ATO) rapidly and strongly reduced TF mRNA. Both drugs also reduced TNF mRNA, but later, and moreover increased IL-1β mRNA. The effect on procoagulant activity of c… Show more

“… 32 IL-1β was also increased after treatment with arsenic trioxide or all- trans retinoic acid. 33 , 34 Similarly, IL-6 displayed opposing effects on cancer cells. 35 For instance, in a phosphatase and tensin homolog (PTEN)-deficient prostate cancer model, IL-6/STAT3 signaling in tumor cells protected against tumor progression.…”

“…Under these conditions, activation of TF is greatly enhanced. TF expression is also increased by inflammatory cytokines, 61 such as present in infection. The activation of TF triggers the coagulation cascade leading to increased thrombin generation and resultant coagulopathic picture described above.…”

“…True DIC (e.g., consumption coagulopathy), which occurs in APL [ 87 , 88 ], is a coagulation (hemorrhagic) disorder, developing due to activation of TF-FVIIa complex-initiated coagulation cascade. In APL, TF is strongly expressed in leukemic promyelocytic cells.…”
