This study was aimed at evaluating the effect of Dihydroartemisinin (DHA)on contractility to drugs in ileum isolated smooth muscle preparations in guinea pigs using in Vitro experimental procedures / standard protocols in an organ bath set up. dihydroartemisinin (1.0 x 10 - 8 - 1.0 x 10 – 5mg/mL) when applied alone and separately excited marked variable effects on guinea pig ileum . In some preparation it showed no response, while in others it produced slight phasic contraction when external calcium (Ca2+) ion was introduced. The slight phasic contractile activity was abolished by verapamil (5x10-3mg/mL). Dihydroartemisinin (1.0 x 10 - 6 - 1.0 x 10 – 5mg/mL) caused marked significant induced contraction – dependent inhibition of acetylcholine, potassium chloride and histamine in depolarizing tyrode solution (p < 0.05).The Inhibitory response maxima of dihydroartemisinin on acetylcholine induced contraction is 17.0±0.1 mm(65.56% inhibition) ; and is moderate when compared to the inhibitory effect of atropine (10-5 M). The result shows that dihydroartemisinin seems to act via non-specific receptor mechanism, with appreciable calcium channel blocking activity and it is safe at therapeutic doses.

Background In malaria-endemic countries, repeated intake of artemisinin-based combination therapies (ACTs) is rampant and driven by drug resistance, improper usage, and easy accessibility. Stress effects and potential liver toxicity due to the frequent therapeutic use of ACTs have not been extensively studied. Here, we investigated the effects of repeated treatment with standard doses of the commonly used ACTs artemether/lumefantrine (A/L) and artesunate-amodiaquine (A/A) on oxidative stress and liver function markers in male mice (BALB/c). Methods Forty Five mice were divided into three groups: control, A/L, and A/A. The drugs were administered three days in a row per week, and the regimen was repeated every two weeks for a total of six cycles. The levels of oxidative stress and liver function markers were measured in both plasma and liver tissue after initial (baseline) and repeated exposures for the second, third, and sixth cycles. Results Exposure to A/L or A/A caused a significant (p < 0.001) increase in plasma malondialdehyde (MDA) levels after the first and repeated exposure periods. However, Hepatic MDA levels increased significantly (p < 0.01) only after the sixth exposure to A/A. Following either single or repeated exposure to A/L or A/A, plasma and liver glutathione peroxidase (GPx) and catalase (CAT) activities, plasma aspartate and alanine transaminase, alkaline phosphatase activity, and bilirubin levels increased, whereas total plasma protein levels decreased significantly (p < 0.001). Varying degrees of hepatocyte degeneration and blood vessel congestion were observed in liver tissues after a single or repeated treatment period. Conclusion Irrespective of single or repeated exposure to therapeutic doses of A/L or A/A, plasma oxidative stress and liver damage were observed. However, long-term repeated A/A exposure can led to hepatic stress. Compensatory processes involving GPx and CAT activities may help reduce the observed stress.

This study investigated the effect of artemether on smooth muscle contractility to drugs in non-pregnant female wistar rats. The effect of artemether (2.1x10 -6 to 2.1x10 -2 ) on isolated uterine and ileum muscle strips was studied using sixteen (16) mature female Wister rats (160 -190g) mounted on organ baths. The study further investigated the effect of artemether on sub-acute histopathological examination of the ileum using standard in vivo procedures in mice models. Data obtained from the study were represented on table, graph and photomicrographs. The results of this study revealed that artemether (2.1x10 -6 to 2.1x10 -2 ) alone did not produce contractile responses, but caused significant inhibition on contractile responses induced by acetylcholine ( 4x10 -5 to 4x10 -1 M) on isolated ileal smooth muscle tissue in rats in a dose dependent manner (p<0.05); also artemether significantly inhibited the contractions induced by oxytocin 8x10 -10 to 1x10 -6 I.U.) and barium chloride ( 8x10 -6 to 8x10 -2 mg/ml ) on uterine smooth muscle strips in rats in a dose dependent manner; with Emax of 13.5 ± 0.2 and 12.6 ±. 0.4 mm respectively, all the observed results were independent of tissue species variation. (p<0.05-0.01). This study revealed that artemether exhibited no adverse effect on the non-pregnant uterus and the rat ileum, hence its safety in the treatment of malaria, as recommended by the World Health Organization.
