Effect of applied and endogenous indol-3-yl-acetic acid on maize root growth

Abstract: The level of endogenous Indol-3-yl-acetic acid (IAA) measured by gas chromatography-mass spectrometry in the elongating zone of intact primary roots of Zea mays showed a good linear correlation with the growth rate of these roots. When they were treated with IAA, their relative elongation decreased; this indicates a supraoptimal content of endogenous IAA. However, the growth of some of the relatively rapidly extending roots was enhanced by such treatment. Interactions between endogenous and applied IAA in the … Show more

“…After chilling on ice, the reaction mixture was diluted with 400 pL of water and the IAA methyl-HFB/[13C6]lAA methyl-HFB was extracted twice with 400 pL of hexane. The hexane phase was washed with 400 pL of water and transferred into a small glass tube, dried under a nitrogen stream, and redissolved in 30 pL of hexane (Pilet and Saugy, 1985).…”

“…The diagnostic ions were at rrdz 278 and 280 corresponding to methyl-ABA and 180-methyl-ABA, monitored in single-ion monitoring (SIM) mode. After methylation, the IAA fractions were derivatized with heptafluorobuturyl imidazole (HFBI) [25]. GC-MS analysis was performed in the electron impact (EI+) mode with the following temperature gradient: 40-95"C (25°C/ min), 95-115°C (5°C/min), 115-300°C (35°C/min).…”

“…Elucidating mechanisms modulating hormone concentrations is therefore critical to our understanding of certain developmental processes. The phytohormone auxin has been associated with deetiolation (OBrien et al, 1985), root initiation and elongation (Pilet and Saugy, 1985;Norcini and Heuser, 1988;Blakesley et al, 1991a), vascular differentiation (Jacobs, 1976), gravitropism and phototropism (Li et al, 1991), endosperm development (Lur and Setter, 19931, somatic embryogenesis (Michalczuk et al, 1992), and fruit ripening (Slovin and Cohen, 1993). Changes in auxin concentration are often associated with these processes, although the coincident or regulatory nature of auxin concentration is not known.…”
