Effect of antithyroid drug on chick embryos during the last week of development: Delayed hatching and decreased cerebellar acetylcholinesterase activity

Abstract: The 'fertilized hen's egg-chick embryo-chick system' is an appropriate animal model for investigating the hypothyroid state during embryogenesis. Decreased cerebellar acetylcholinesterase activity after MMI treatment was assumed to relate to a mechanism of motor and cognitive deficits in congenital hypothyroidism.

“…Moreover, the hatching rate was strikingly low in the MMI-treated group and this was deemed significant, not only statistically, but also physiologically. This result is in line with the observation that chicken embryos have been reported to have delayed hatching upon exposure to MMI (Haba et al, 2011). Interestingly, another compound shown to affect zebrafish hatching rate is perfluorooctanoic acid (PFOA), an established thyroid-active compound (lopez-espinosa et al, 2012;Melzer et al, 2010;Zheng et al, 2011).…”

“…Likewise, altering levels of SF1, NR5A2, ESRRβ, and several members of the SMAD and LHX families were also detected in the chick ovaries during embryonic to post-hatching transition (Oreal et al, 2002;Ghafari et al, 2007;Carre et al, 2011). In contrast, information about the functions of NF1, THRβ, and the TEAD family remains scarce in the avian ovary, all of which were evidenced to be involved in regulating cell survival, proliferation, and adhesion partially by acting as components of multiple signal transduction (e.g., the Hippo and Ras-MEK/Akt) pathways and some of which were expressed in a variety of tissues of the chick embryos (Schafer et al, 1993;Cao et al, 2008;Haba et al, 2011). Significantly, expression of these transcription factors in embryonic and post-hatching geese ovaries was determined by RNA-seq and qRT-PCR.…”

“…Hatching is delayed or even inhibited in embryos treated with different TH inhibitors. Furthermore, an administration of TH into the egg retracts the yolk sac and shortens the incubation time [ 18 , 33 , 34 , 35 , 36 , 37 ]. Based on findings from earlier studies it is therefore likely that the difference in incubation time seen in the TSHR chickens is caused by altered TH levels due to the mutation in the TSHR d/d genotype.…”
