Effect of amino acid substitutions in conserved residues in the leader peptide on biosynthesis of the lantibiotic mutacin II

Abstract: The lantibiotic mutacin II, produced by Streptococcus mutans T8, is a ribosomally synthesized peptide antibiotic that contains thioether amino acids such as lanthionine and methyllanthionine as a result of post-translational modifications. The mutacin II leader peptide sequence shares a number of identical amino acid residues with class AII lantibiotic leader peptides. To study the role of these conservative residues in the production of active antimicrobial mutacin, 15 mutations were generated by site-directe… Show more

“…3), including streptococcin A-FF22 (SA-FF22), lacticin 481, variacin (Pridmore et al, 1996), mutacin II (Woodruff et al, 1998) and salivaricin A (Ross et al, 1993). It contains a GG (double-glycine) motif immediately preceding the cleavage site, and a conserved EL sequence (Sablon et al, 2000;Chen et al, 2001). The leader peptide of bovicin HJ50 contains a TVS motif instead of the conserved EVT/EVS sequences.…”

“…It may act in a protective role for the producing organism, as its removal is required for bioactivity (10)(11)(12)(13)(14)(15). The leader peptide could also serve as a recognition motif for the transport machinery because non-lantibiotic peptides attached to leader peptides are secreted (16,17), or as a scaffold for the lantibiotic synthetases (11,(18)(19)(20).…”

“…With the exception of a very recent report on ComC (27), these previous studies focused mainly on characterization of the protease with investigation of substrate recognition receiving little attention. The study of lantibiotic proteases has been restricted to limited in vivo mutagenesis of the leader peptides (10,13,15,28,29).…”
