Effect of Alliin on Vascular Functions

Physicochemical characteristics of domestic garlic by varieties and their quality changes during aging of black garlic

In this study, the quality characteristics of the representative domestic garlic varieties in Korea (Namdo, Daeseo, Hongsan, and Uiseong garlic) were compared and their changes according to the aging process of black garlic were investigated. Each garlic sample was aged for 15 days according to the manufacturing process of black garlic. The total phenolic compound contents were 85.2-109.7 mg GAE/100 g in raw garlic samples, which increased with the aging period, and Hongsan garlic showed the highest content in both raw and aged black garlic. The content of alliin was 848.3-1,087.5 mg/100 g in raw garlic samples, and it decreased significantly on the 5th day (7.3-20.6 mg/100 g) in aged garlic. The contents of GSAC (γ-glutamyl-S-allyl-cysteine) and GS1PC (γ-glutamyl-S-1-propenyl-l-cysteine) in raw garlic samples were 202.1-541.0 mg/100 g and 311.1-474.3 mg/100 g, respectively, but gradually decreased during the aging process. On the other hand, the SAC (S-allyl-cysteine) and S1PC (S-1-propenyl-l-cysteine) contents of raw garlic were very low in all four varieties, but they were highest on the 5th day of aging and tended to decrease thereafter. On the 5th day of aging, the SAC content of Namdo garlic (208.0 mg/100 g) was highest (p<0.05), and S1PC content was significantly higher in Uiseong garlic (66.2 mg/100 g) than the others.

Platycodon grandiflorum A. De Candolle (Korean name, 'Doraji') is a perennial plant containing various triterpenoid saponins. The roots of this plant have traditionally been used as a food material in Korea. Here, we prepared a fermented P. grandiflorum extract (PG). Although it was previously reported that P. grandiflorum A. extract has a variety of physiological functionalities, including anti-inflammatory and anti-oxidant activities, little is known about its vascular functions. In this study, we executed a series of experiments to identify the effect of PG on endothelial cells. PG at a high concentration (100 μg/ml) was found to induce cell detachment, whereas PG at a low concentration (0.1 μg/ml) appeared to promote cell proliferation and migration in bovine aortic endothelial cells. The cell detachment induced by the high concentration was not associated with cell death, such as apoptosis, necrosis, and autophagy. In addition, we found that PG at the high concentration formed a small vesicular structure called an endothelial microparticle (EMP). The EMP was prepared by centrifugal fractionation and determined with flow cytometry and a microscope. Interestingly, PG-induced cell detachment was found to be mediated by EMP. We furthermore determined that PG at the low concentration activated Akt, a crucial cell-signaling molecule, and then controlled cell proliferation and migration. Overall, our findings suggest that PG at low doses maintains vascular stability by promoting endothelial cell proliferation, and enhances the efficacy of wound healing by cell proliferation and migration activity.

-This study is a basic study on the development of functional substances involved in obesity prevention, lipid metabolism, and immune regulation. Male Sprague-Dawley rats were fed a high-fat diet for 10 weeks. Allium monanthum extracts (AME) were administered orally to obesity-induced rats, and their lipid-lowering, antioxidative and various types of biological effects related to the immune system were examined. Blood free fatty acid and triglyceride concentrations decreased as the dose of AME increased. Total cholesterol and LDL cholesterol concentrations in the blood decreased as the dose of AME increased. The total cholesterol concentrations in the liver of the AME-treated groups were lower than the control group. The thiobarbituric acid reactive concentrations were lower in the plasma and liver of all AME-treated groups than the control group. Plasma AST and ALT activities did not show any significant differences among the treatment groups. IL-1β and IL-6 concentrations in the liver tended to decrease as the dose of AME increased. TNF-α and IL-10 concentrations did now show any significant differences compared to the control group. Lower expression levels of TNF-α, Apo-B and Apo-E genes were found in the AME-treated groups. Taken together, these results indicate that AME may show positive effects in lipid lowering, antioxidation and anti-inflammation.
