Effect of aging on airway remodeling and muscarinic receptors in a murine acute asthma model

Abstract: Background and objectivesThe influence of aging on the development of asthma has not been studied thoroughly. The aim of this study was to investigate age-related airway responses involving lung histology and expression of muscarinic receptors in a murine model of acute asthma.MethodsFemale BALB/c mice at the ages of 6 weeks and 6, 9, and 12 months were sensitized and challenged with ovalbumin (OVA) for 1 month (n = 8–12 per group). We analyzed inflammatory cells and T-helper (Th)2 cytokines in bronchoalveolar… Show more

“…Cell senescence in the lung is closely related to the pathogenesis of asthma. The senescence of airway epithelial cells is associated with the development of asthma, such as airway remodeling . Thus, our results strongly suggest that senescence of airway epithelial cells after ITGB4 deficiency may have a close relationship to the pathogenesis of asthma.…”

“…45 In an aging model of asthma, 6-month-old mice had more prominent collagen accumulation and airway smooth muscle hypertrophy in their airways than younger mice. 46 Age-related alteration in collagen synthesis and degradation and asthmaspecific airway inflammation can synergistically contribute to the progression of airway remodeling in elderly patients with asthma.…”

“…The hyperplasia of the goblet cells in the epithelial lining was expressed by a score according to the percentage of the goblet cells in the epithelial cells. Five-point scoring system (grade 0-4) was used as follows: no goblet cell (grade 0), <25% (grade 1), 25-50% (grade 2), 51-75% (grade 3), and >75% (grade 4) [21,22]. The preparations, in which the maximum internal diameter of the bronchioles were two fold or greater as large as the minimum internal diameter, were not used for analysis.…”
