Effect of aerobic exercise on acquired gefitinib resistance in lung adenocarcinoma

Abstract: Highlights Long-term gefitinib induction could increase the level of oxidative stress in lung adenocarcinoma cells and reduce the antioxidant capacity. Acquired resistance to gefitinib in lung adenocarcinoma was closely related to the high expression of HIF-1 and ALDH1 and the enrichment of CSCs. The inhibitory effect of aerobic exercise on oxidative stress can effectively reduce the expression of HIF-1 and ALDH1 and inhibit the enrichment of CSCs, which c… Show more

“…ESCC patients with Pg-positive infection have shorter survival and poor chemotherapy effect . In 2023, it was found that Pg could colonize the cytoplasm of ESCC cells, induce cancer cells to overexpress GSK3β, enhance their mitochondrial function, reduce their sensitivity to chemotherapy drugs, and promote their malignant evolution. , Therefore, it is essential to explore the molecular mechanism of Pg pathogenesis in ESCC for improving treatment and patient prognosis.…”

“…The percentage of ALDH1 + CSCs in each group was determined by Flow Cytometry (Beckman, USA). The difference in the percentage of ALDH1 + CSCs between the above two tubes was the final percentage of CSCs. , …”

“…After rewarming, secondary antibody (China Kangwei Biological Technology Co., 1:2000 TBST dilution) was added and incubated for 2 h. An ECL luminescent developer (Invitrogen Co. of America) was used to detect the target protein bands, a gel imaging system (BIO-RAD Co. of America) was used to collect images, and Image Lab software was used to measure the grey value of the protein bands. The grey value ratio between the target protein and the reference protein bands was used as the final relative protein expression [ 22–23 ]. The experiment was repeated three times.…”

“…Standard drugs for lung cancer treatment, cisplatin/gemcitabine,and menadione, reduced ALDH1 expression ( 68 ), while the elimination of ALDH1A1 significantly increased apoptosis and decreased resistance to cisplatin ( 69 ). Fat1 overexpression ( 70 ), aerobic exercise ( 71 ), nilotinib, erlotinib ( 72 ), all-trans retinoic acid ( 73 ), itraconazole ( 74 ), cryptotanshinone ( 55 ), CFTR31 ( 75 ), fluorescent interleukin ( 76 ), pomegranate ( 77 ), ginsenoside Rg3 ( 78 ), puerarin 6″-O-xyloside ( 79 ), glycolysis inhibitor PFK158 ( 67 ), globulin ( 80 ) can reduce expression of ALDH1 and drug resistance in lung cancer, mainly involved NF-κB pathway, Wnt pathway, Src-STAT3 signaling axis, Wnt/β-catenin/STAT3 axis, Akt/c-Myc signaling pathway. Therefore, in the treatment of lung cancer, targeting ALDH1 has broad prospects and should be further studied and explored.…”
