Flow and Thixotropic Parameters for Rheological Characterization of Hydrogels

The goal of this paper was to design several sodium carboxymethylcellulose hydrogels containing a BCS class II model drug and to evaluate their flow and thixotropic properties. The rheological measurements were performed at two temperatures (23 °C and 37 °C), using a rotational viscometer. The hydrogels were stirred at different time intervals (10 s, 2, 5, 10 and 20 min at 23 °C, and 10 s, 2 and 5 min at 37 °C), with a maximum rotational speed of 60 rpm, and the corresponding forward and backward rheograms were recorded as shear stress vs. shear rate. For all hydrogels, the rheological data obtained at both temperatures showed a decrease of viscosity with the increase of the shear rate, highlighting a pseudoplastic behaviour. The flow profiles viscosity vs. shear rate were quantified through power law model, meanwhile the flow curves shear stress vs. shear rate were assessed by applying the Herschel-Bulkley model. The thixotropic character was evaluated through different descriptors: thixotropic area, thixotropic index, thixotropic constant and destructuration thixotropic coefficient. The gel-forming polymer concentration and the rheological experiments temperature significantly influence the flow and thixotropic parameters values of the designed hydrogels. The rheological characteristics described have an impact on the drug release microenvironment and determine the stasis time at the application site.

The powdered form of itraconazole is white, flavorless, and stable, but it dissolves slowly in water. In comparison to palmitic acid, stearic acid has a stronger affinity (2.57), as shown by the partition coefficient test. UV-visible spectra analysis reveals critical compound insights. Fourier-transform infrared (FTIR) identifies significant peaks in itraconazole and related compounds. tween 80 is selected for solid lipid nanoparticles (SLN) based on their appearance. Optimization using 0.5% w/v tween 80 yielded better particle capture. Thermal analyses display distinctive characteristics of itraconazole, palmitic acid, and polyvinyl alcohol. Viscosity studies mirror tear film behavior. Transmission electron microscope (TEM) images demonstrate SLN’s potential ocular application without irritation. Antifungal studies exhibit effective inhibition of Candida albicans and Aspergillus flavus growth. SN-7 reveals strong encapsulation efficiency, mean vesicle size, and sustained drug release over 12 hours. Pharmacokinetic studies shed light on SLNs’ influence on drug absorption and distribution, indicative of potential in drug delivery systems. These findings offer promising prospects in formulating enhanced drug delivery mechanisms and understanding their impact on drug pharmacokinetics. High-performance liquid chromatography (HPLC) analysis confirmed SN-7’s singular peak at 6.59 minutes. Both formulations showcased skin penetration and stability over three months under varied conditions. Skin irritation tests on rats indicated minimal reactions, affirming low irritancy, which is crucial for patient comfort and acceptance in topical drug delivery. These assessments ensure safety and effectiveness in treating fungal infections.

The Preformulation study of Luliconazole elucidated its properties and optimization for a self-Nanoemulsifying drug delivery system (SNEDDS). Luliconazole, a tasteless, odorless white powder, exhibited diverse crystalline forms with a melting point of 153.6°C. Notably, it demonstrated high solubility in Oleic Acid and Isopropyl Myristate, corroborated by partition coefficients of 2.80 in n-octanol. UV spectroscopy validated its reliability for analysis, exhibiting a linear standard curve (Y = 0.1013x) with R2 = 0.9988 at 261 nm. The FTIR spectra of Luliconazole and excipients (Isopropyl Myristate, Oleic Acid, Tween 80, and PEG 400) highlighted compatibility crucial for SNEDDS formulation, showcasing distinctive peaks at specific wave numbers. A 32-factorial design optimized IS5 batches, enhancing efficacy and stability through water dilutions (1:1). Thermal analysis, including DSC and XRD, revealed distinct thermal behaviors and crystalline structures, crucial for understanding formulation interactions. The SNEDDS exhibited increased viscosity due to the surfactant, promoting prolonged surface retention and demonstrating significant inhibition of Candida albicans growth, quantified in Table 12 and Fig. 21. In vitro release testing displayed sustained drug liberation in STF (pH 7.4), showcasing diverse kinetics. Entrapment efficiency in IS-4 reached 74 ± 0.8%, ensuring stability (1.54 ± 0.008) vital for emulsification and drug release behavior. HPLC analysis in an ex-vivo permeation study identified a single peak at 6.59 minutes in IS-5, showcasing skin permeation and bloodstream entry, indicating potential bioavailability for fungal infections. The stability study confirmed IS-5's consistent effectiveness, purity, and potency across varied conditions (4°C, 25°C, 40°C; 15%, 65%, 75% RH), crucial for ensuring a reliable and enduring product. Additionally, skin irritation studies validated IS-5's safety for transdermal drug delivery, essential for patient comfort in pharmaceutical applications.
