Efeitos citotóxicos de anestesia local com lidocaína/ropivacaína em linhagens celulares de melanoma humano

Abstract: LAs have cytotoxic activity on human melanoma cell lines in a time-, concentration- and agent-dependant manner. Apoptosis in the cell lines was mediated through activity of caspases-3 and caspases-8.

“…Though the high concentration of plain ropivacaine solution showed a higher anesthetic score than low dosage, the highest concentration of 1% Ropivacaine HCl Injection (8.3 mg/mL of ropivacaine base) available in clinic is almost equal to the concentration of low dosage in the present study. Some evidence suggests that the cell toxicity effect of local anesthetics may be determined in a concentration-dependent manner from in vitro testing with specific cell lines, such as Schwann cells,17 human melanoma cells18 or rabbit intervertebral disc cells 19. A comparative advantage of using a sustained-release formulation is it will decrease the local concentration of un-bound anesthetic by holding it in reserve in MVR to reduce the local toxicity.…”

“…In addition, they reveal the potential of LDC for the treatment of melanoma, in association with an antineoplastic agent. Previous studies pointed out the anticarcinogenic effect of LDC in different tumor models, such as breast [68], bladder [20], squamous and basal cell carcinoma [69], gastric cancer [28] and melanoma cell [19]. A recent review shows that LDC acts as a chemosensitizer to other antineoplastic agents, inhibiting the growth of tumors after single use at different concentrations through the regulation of epigenetic modifications, promotion of pro-apoptotic pathways and angiogenic inhibitor.…”

“…Recently, several works have described the adjuvant anticancer action of local anesthetics such as LDC [19,20], bupivacaine [21][22][23][24] and ropivacaine [25]. LDC can suppress cancer cell growth (in vitro and in vivo) through several mechanisms: regulation of epigenetic changes, promotion of pro-apoptotic pathways and regulation of ABC transporters, also preventing metastasis and angiogenesis.…”
