A nutrient medium was selected for the efficient production of exopolysaccharide (EPS) by A. pullulans C7 strain. The production of pullulan polysaccharide was evaluated on nutrient media with traditional carbon sources and cheap substrates that were plant wastes. For maximum EPS accumulation, we proposed an optimized Czapek-Dox medium with glucose as a carbon source, sodium nitrate as a nitrogen source, and C/N=232:1 ratio (EPS yield 12.79±0.64 g/l). Medium with grape pomace 5% (EPS yield was 15.08±0.34 g/l) and medium with topinambour tuber hydrolysate 5% (EPS yield was 14.44±0.21 g/l) was proposed as a cheap substrate. Edible films with antimicrobial activity were obtained on the basis of the isolated polysaccharide. The antibacterial activity of films against Escherichia coli 603 and Staphylococcus aureus ST228 was shown when essential oils of rosemary (zones of growth inhibition from 8.41±0.71 to 9.98±0.32 mm) and oregano (zones of growth inhibition from 8.09±0.51 to 9.54±0.24 mm) were added to pullulan. The addition of xanthan gum and glycerol to the films increased their strength and elasticity. The infrared spectrum of the pullulan film showed absorption bands characteristic of polysaccharide structures.

The paper studies the biological properties of film coatings created on the basis of polysaccharides (xanthan and carboxymethylcellulose) for use in various branches of the agro-industrial complex. It was found that samples of these film coatings completely decomposed in the soil in 7 days, and did in the aquatic environment in an average of 3.5 h. The effect of these film coatings on the organism of laboratory animals (rats) was studied by adding them to animal feed (experimental group). During the experiment, no deviations in behavior were noted in the experimental group of rats; no death in both the control and experimental groups of animals occurred. The studied film coating had no negative effect on the large intestine microbiocenosis and the animals’ weight. It was shown that the number of mesophilic aerobic and facultative anaerobic microorganisms in the experimental group was 3 times less than in the control one, and the number of lactic acid bacteria of the genus Lactobacillus was 3.3 times higher than in the control group. The results obtained indicate the biological safety of the use of the studied film coatings.

This work describes the design of a film coating based on polysaccharides of bacterial and plant origin with several methods of application (by spraying, brushing and wrapping) on food. We used the bacterial polysaccharide xanthan as a thickener, the plant polysaccharide carboxymethyl cellulose (CMC) as a structurant, and the biological additive lecithin as a plasticizer. Depending on the film coating application method, concentrations of the studied polymers were selected with the same component composition. A technology for preparing the film coating was developed. The influence of heating (40, 50, 60, and 70 ° C) on the solubility of polysaccharide solutions with several component ratios was studied; the temperature of 50 ° C was chosen as resulted in a strong, stable, but not too viscous gel. The effect of concentration on the appearance and thickness of the film coatings was studied.
