Edematous severe acute malnutrition is characterized by hypomethylation of DNA

Abstract: Edematous severe acute childhood malnutrition (edematous SAM or ESAM), which includes kwashiorkor, presents with more overt multi-organ dysfunction than non-edematous SAM (NESAM). Reduced concentrations and methyl-flux of methionine in 1-carbon metabolism have been reported in acute, but not recovered, ESAM, suggesting downstream DNA methylation changes could be relevant to differences in SAM pathogenesis. Here, we assess genome-wide DNA methylation in buccal cells of 309 SAM children using the 450 K microarra… Show more

“…There is increasing interest in whether perturbations in the gut microbiome underlie fatty liver changes and generation of edema ( 47 ). Those with edema also have differences in DNA methylation compared with those without edema, but the significance of these epigenetic changes or the patterns of genes involved is unclear ( 48 ). The mixed clinical presentation of marasmic-kwashiorkor was shown by Irena et al ( 16 ) to have a higher mortality than kwashiorkor alone, suggesting that the combination of low WHZ and edema may increase mortality risk, but further studies from other settings are needed to clarify the outcomes of children with and without edema.…”

“…As an additional quality measure after the filtering mentioned above, only the probes having a detection p < .0001 versus the background in more than 90% of the samples 43 were used for the differential methylation analysis. A minimum absolute difference (delta of beta) of 10% was considered relevant.…”

“… 5 Undernourished cohort from Bangladesh captured differential histone methylation in peripheral blood, 6 and cohorts from Jamaica and Malawi characterized differential DNA methylation. 7 However, epigenetic and transcriptional drivers of EED pathogenesis in the affected small intestine and longitudinal biomarkers to predict growth have not yet been defined.…”
