In the bone tissue engineering field, there is a growing interest in the application of bioactive glass scaffolds (45S5Bioglass(®)) due to their bone bonding ability, osteoconductivity and osteoinductivity. However, such scaffolds still lack some of the required functionalities to enable the successful formation of new bone, e.g. effective antibacterial properties. A large number of studies suggest that selenium (Se) has significant role in antioxidant protection, enhanced immune surveillance and modulation of cell proliferation. Selenium nanoparticles (SeNp) have also been reported to possess antibacterial as well as antiviral activities. In this investigation, uniform, stable, amorphous SeNp have been synthesized and additionally immobilized within spherical PLGA particles (PLGA/SeNp). These particles were used to coat bioactive glass-based scaffolds synthesized by the foam replica method. Samples were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS) and transmission electron microscopy (TEM). SeNp, 45S5Bioglass(®)/SeNp and 45S5Bioglass(®)/PLGA/SeNp showed a considerable antibacterial activity against Gram positive bacteria, Staphylococcus aureus and Staphylococcus epidermidis, one of the main causative agents of orthopedic infections. The functionalized Se-coated bioactive glass scaffolds represent a new family of bioactive, antibacterial scaffolds for bone tissue engineering applications.

Alkoxysilane precursors are the most widely used silica source for sol-gel preparation of silicate-based bioactive glass. However, due to their high cost, alternative sources such as bentonite clay are desirable. In the present work, bentonite clay was reacted with sodium hydroxide (NaOH) to extract sodium metasilicate (Na 2 SiO 3 ). The obtained Na 2 SiO 3 was converted to gel which was then sintered at 950 ℃ for 3 h to give the bioactive glass in the quaternary composition SiO 2 -NaO-CaO-P 2 O 5 . The resulting glass was incubated in simulated body fluid (SBF) for 0-7 days to evaluate the bioactivity. Furthermore, glass samples were characterized before and after SBF study by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR). Results obtained showed the presence of Na 2 Ca 2 Si 3 O 9 (combeite) crystal as the major crystalline phase and the formation of hydroxyapatite (HA) and hydroxycarbonated apatite (HCA) on the surface of the glass after immersion in SBF. The material showed potentials for application as scaffold in bone tissue repair.

A comprehensive study on compositional and structural changes in 45S5 bioglass products exposed to simulated body fluid

The interaction of fully dense 45S5-bioglass derived samples produced by Spark Plasma Sintering (SPS) with simulated body fluid (SBF) solution was investigated in detail taking advantage of the Rietveld refinement method to quantitatively evidence the corresponding microstructural and compositional changes. It is observed that, when the original amorphous nature is mostly (75 wt%) preserved in the material during sintering (550°C, 2 minutes), the resulting specimens dissolve faster and determine relatively higher pH increase and ions release in the SBF solution. Correspondingly, a relatively lower amount of hydroxycarbonate apatite (HCA) is formed on their surface. In contrast, a more extensive apatite layer with trabecular structure is generated within 3 days storage on the surface of fully crystallized samples obtained at 600°C by SPS, which only consists of Na-Ca silicate grains (20 nm). Moreover, as the sintering temperature and dwell time were increased to 700°C and 20 minutes, respectively, a rhenanite-like phase was also formed (about 15 wt%), other than crystallites growth to 90 nm. Interestingly, the presence of rhenanite provides a beneficial contribution for the production of the HCA layer, which was found the largest for this system when considering storage periods of 7 and 14 days, respectively.
