Spawning sites play a key role in the reproduction of fish allowing populations to endure over time. The Nechí River is an important spawning area for potamodromous fish species where one of the threats is dam construction. In order to determine the importance of the Nechí River as a spawning site in the Magdalena River basin, sampling was conducted during the low-water-to-high-water season transition period between 2018 and 2019 at seven sampling sites. The average density of ichthyoplankton was 42.4 ind.10m-3 (SD = 7.1). Of the individuals in the post-larval stage, seven migratory species were identified, and two additional taxa were identified to genus; Prochilodus magdalenae, Megaleporinus muyscorum, and Pseudoplatystoma magdaleniatum presented the greatest density. At the temporal level, the greatest density of larvae of potamodromous species was observed in the first high-water season of 2019 with a total of 5.7 ind.10m-3(SD = 1.044), of which the most representative at the seasonal level were the Cauca River, Magdalena River, and Nechí River before it flows into the Cauca River. There were significant differences in the frequency of embryos and vitelline larvae of the potamodromous species in the interaction of the sampling sites and high-water seasons, as well as with the density of post-larvae. The average drift distance of the spawning areas is roughly 52.1 km. A positive association was found between the volume of turbined water and the presence of ichthyoplankton in the Porce River site, after discharge from the Porce III Hydroelectric Plant. The Nechí River is an important spawning site and there seems to be an association between the increase in ichthyoplankton densities and the distance to the dam (Porce III) as long as there are floodplains along the course of the river.

Habitat fragmentation is among the main threats to freshwater fish biodiversity, with expected effects including genetic impacts due to disturbance of migration and declining population size. Major concern falls on highly exploited species such as Prochilodus magdalenae, a migratory characiform fish endemic to the Magdalena River basin supporting much of the Colombian artisanal fishery, whose migration route was interrupted by the Ituango hydroelectric project in the Cauca River. To determine the potential effects of fragmentation, this study evaluated the population genetics of this species on both geographical (upstream and downstream of the dam) and temporal (before and after the construction) scales by using 11 species-specific microsatellite markers. Contrary to expectation, genetic diversity and structure remained relatively stable. This study provided no evidence of genetic impacts on P. magdalenae associated with fragmentation over the short term (4–10 years, 5–12 generations) despite persisting threats, genetic evidence of bottleneck, and a high degree of inbreeding, showing the ability of this species to withstand disturbance of its habitat.

The adaptative responses and divergent evolution shown in the environments habited by the Cichlidae family allow to understand different biological properties, including fish genetic diversity and structure studies. In a zone that has been historically submitted to different anthropogenic pressures, this study assessed the genetic diversity and population structure of cichlid Caquetaia kraussii, a sedentary species with parental care that has a significant ecological role for its contribution to redistribution and maintenance of sedimentologic processes in its distribution area. This study developed de novo 16 highly polymorphic species-specific microsatellite loci that allowed the estimation of the genetic diversity and differentiation in 319 individuals from natural populations in the area influenced by the Ituango hydroelectric project in the Colombian Cauca River. Caquetaia kraussii exhibits high genetic diversity levels (Ho: 0.562–0.885; He: 0.583–0.884) in relation to the average neotropical cichlids and a three group-spatial structure: two natural groups upstream and downstream the Nechí River mouth, and one group of individuals with high relatedness degree, possibly independently formed by founder effect in the dam zone. The three genetic groups show recent bottlenecks, but only the two natural groups have effective population size that suggest their long-term permanence. The information generated is relevant not only for management programs and species conservation purposes, but also for broadening the available knowledge on the factors influencing neotropical cichlids population genetics.
