EBS is a bivalent histone reader that regulates floral phase transition in Arabidopsis

Abstract: The ability of cells to perceive and translate versatile cues into differential chromatin and transcriptional states is critical for many biological processes. In plants, timely transition to a flowering state is crucial for successful reproduction. EARLY BOLTING IN SHORT DAY (EBS) is a negative transcriptional regulator that prevents premature flowering in Arabidopsis thaliana. We found that EBS contains bivalent bromo-adjacent homology (BAH)-plant homeodomain (PHD) reader modules that bind H3K27me3 and H3K4m… Show more

“…These histone marks are catalyzed and removed by specific enzyme complexes that are referred to as the writers and erasers, and are recognized by reader proteins (Liu et al 2010). For example, H3K27me3, which is a representative repressive marker that exhibits high deposition on many stress‐responsive genes, is catalyzed by the polycomb repressive complex 2 (PRC2) via the histone methyltransferases CURLY LEAF (CLF), SWINGER (SWN), and MEDEA (MEA) and their cofactors; meanwhile H3K27me3 is recognized by the PRC1 complex via the H3K27me3 reader proteins LIKE HETEROCHROMATIN PROTEIN 1 (LHP1), EARLY BOLTING IN SHORT DAY (EBS), and SHORT LIFE (SHL) and their cofactors in Arabidopsis (Mozgova and Hennig 2015; Li et al 2018; Yang et al 2018b). As an active marker, H3K4me3 is also present on a large number of stress‐responsive genes.…”

“…The extent of the early flowering phenotype in long-day photoperiods varies across greenhouses (Zemach et al, 2013;Kang et al, 2015). Therefore, we included additional genotypes to control for this effect (ebs1 flowers early and hda6 flowers late; Yu et al, 2011;López-González et al, 2014;Yang et al, 2018). All mutants display a dwarf stature, with corresponding mass reductions of the fifth rosette leaf (means ranged 12.2 to 19.4 mg) relative to Col-0 (mean of 33.5 mg; n = 8, P , 0.001; Fig.…”

“…While plants and animals utilize distinct sets of accessory proteins to recognize methylated H3K27 (10,(16)(17)(18), they are generally thought to mediate repression in the context of a canonical PRC1 complex (7,(19)(20)(21). In fungal lineages that employ H3K27 methylation as a repressive chromatin mark, however, core PRC1 components are notably absent (7).…”
