Members of the Meis family of TALE homeobox transcription factors are involved in many processes of vertebrate development and morphogenesis, showing extremely complex transcriptional and spatiotemporal expression patterns. In this work, we performed a comprehensive study of chicken Meis genes using multiple approaches. First, we assessed whether the chicken genome contains a Meis3 ortholog or harbors only two Meis genes; we gathered several lines of evidence pointing to a specific loss of the Meis3 ortholog in an early ancestor of birds. Next, we studied the transcriptional diversity generated from chicken Meis genes through alternative splicing during development. Finally, we performed a detailed analysis of chick Meis1/ 2 expression patterns during early embryogenesis and organogenesis. We show that the expression of both Meis genes begins at the gastrulation stage in the three embryonic layers, presenting highly dynamic patterns with overlapping as well as distinct expression domains throughout development. PAX6 expression was examined during early development of the diencephalon and midbrain of Alligator using an immunocytochemical methodology. These observations focused on the basal plate to determine whether diencephalic prosomere organization in this region followed a pattern previously identified for alar plate areas. PAX6 expression was also described in alar diencephalic regions and the adjacent midbrain. PAX6 (+) cells in the basal plate were first seen in prosomere 1 at stage 7, in the midbrain at stage 10, and lastly in prosomeres 2 and 3 at stage 11. By stage 12, a nearly continuous column of PAX6 (+) cells extended from the midbrain basal plate through the entire diencephalon. In the diencephalon, PAX6 (+) cells in the basal plate were of greatest number in prosomere 1, least in prosomere 2, and intermediate in prosomere 3. This pattern of PAX6 expression distinguished these individual basal plate prosomeres. These results indicate that basal plate prosomeres follow a pattern similar to alar plate prosomereric organization during the later stages of early diencephalon development. Over a comparable time period of early diencephalon development, similar observations have been made in chick basal plate. In Alligator and chick, PAX6 expression in the basal plate is similar in the midbrain and prosomere 1 but different in prosomeres 2 and 3: present in Alligator and absent in chick. In alar plate areas of the Alligator diencephalon, PAX6 expression follows a similar pattern to that described for chick and mouse. These similarities in PAX6 expression in alar diencephalic prosomeres suggest that this is a common feature of amniotes. Differential PAX6 expression in alar prosomere 1 and the midbrain in Alligator is similar to that described for a wide range of species which suggests that these features are common to all vertebrates.

Roughly 45 years ago, when I started teaching neuroanatomy, it was customary to present the brainstem as built by three blocks, the medulla oblongata, the pons (with the cerebellum) and the midbrain. The ponto-mesencephalic or caudal midbrain limit was traced along the sulcus that runs above the pons and its continuation, the middle cerebellar peduncle. The boundary passed below the lemniscal trigone (which contains the superior cerebellar peduncle and the lateral lemniscus tracts), and finished dorsally at the superior medullary velum, caudal to the trochlear nerve root. The mes-diencephalic border represented the rostral midbrain limit, which was given by an imaginary plane passing in front of the superior colliculus (behind, across, or in front of the posterior commissure, depending on the authors). This limit passed behind the medial geniculate body and ended ventrally at the upper end of the interventricular fossa, close to the mamillary bodies. The whole interpeduncular fossa and visible peduncles were held to be mesencephalic, and so were the oculomotor and trochlear nerves (Figure 1a).This classic concept of the midbrain limits has not stood the test of time, due to accrued developmental knowledge and recent gene expression patterns implying strongly that both limits defined above are inexact. It has been shown that the old 'midbrain' does not represent a developmental unit, because it is too inclusive: it encompasses diencephalic derivatives rostrally and hindbrain derivatives caudally. A new, more restricted concept of the midbrain has emerged (Figure 1b), which is consistent with gene patterns, patterning mechanisms, and modern notions about neuromeric structure of the neural tube (the prosomeric model). This essay aims to summarize the new data and explain the resulting changes in the midbrain concept, ending with a summary of present knowledge about its inner structure (internal transversal, longitudinal and radial parts, and corresponding derivatives). Molecular Patterning of the MidbrainIt seems convenient to summarize at the start the present embryologic knowledge about patterning of the midbrain, since this provides a causal background that necessarily applies to any morphologic considerations. Incipient differential specification of the neural ectoderm occurs along the dorsoventral and anteroposterior dimensions, starting at early neural plate stages.
