Early Onset of Spontaneous Renal Preneoplastic and Neoplastic Lesions in Young Conventional Rats in Toxicity Studies

Abstract: Although occurring in aged laboratory rodents, spontaneous renal tumour are unknown in animals younger than 18 weeks. A survey on renal preneoplastic and neoplastic lesions has been performed on Sprague-Dawley rats from general toxicology studies over the period January 2004 - May 2006. Data from 2249 rats necropsied and 1206 rats (688 males and 518 females) examined microscopically from 52 studies, are reported. The age range at necropsy was between 12 and 18 weeks and all the animals were obtained from the s… Show more

“…In young adult rats (studies up to 26 weeks), the data on tumor incidences are limited. However, there are several case reports or organ-related summaries published (e.g., Chang et al 2008; Crabbs et al 2013; Matsushima et al 2010; Reindel et al 1996; Shiraki et al 2012; Tsunenari, Yamate, and Sakuma 1997), and summary data are available in the literature (e.g., Ikezaki, Takagi, and Tamura 2011; Lanzoni et al 2007; Son et al 2010). Tumors may be encountered in animals as young as 9 weeks.…”

“…All macroscopic and microscopic findings (including a renal carcinoma in one mid dose female) were ruled incidental and unrelated to treatment. Renal tubular tumors have been observed sporadically in Sprague-Dawley rats of similar age [11] , [15] .…”

“…In the case of AV tumors, very little information is available regarding their occurrence in SD rats. Lanzoni et al (2007) conducted a survey in young ( 18 weeks of age) SD rats from toxicity studies of shorter duration, reporting an overall incidence of 0.66% for preneoplastic and neoplastic lesions in the kidney; however, this study did not recognize AV tumors as a distinct entity and included foci of atypical hyperplasia. Females were affected more commonly than males (0.97% and 0.44%, respectively).…”
