Early life infection and proinflammatory, atherogenic metabolomic and lipidomic profiles in infancy: a population-based cohort study

Abstract: Background:The risk of adult onset cardiovascular and metabolic (cardiometabolic) disease accrues from early life. Infection is ubiquitous in infancy and induces inflammation, a key cardiometabolic risk factor, but the relationship between infection, inflammation, and metabolic profiles in early childhood remains unexplored. We investigated relationships between infection and plasma metabolomic and lipidomic profiles at age 6 and 12 months, and mediation of these associations by inflammation. Methods:Matched in… Show more

“…Due to the elevated risk of mortality with these diseases, there is a need for strategies in its identification, prevention, and treatment within healthcare. In 2022, as evidenced in Figure 5, the bulk of research articles focussed on related diseases: CVD, 232–255 diabetes, 166,233,249,250,256–278 liver disease, 51,279–293 MetS 277,294,295 and obesity 277,295–305 …”

“…In another recent large prospective cohort study, metabolomic and lipidomic profiles and their relationship to infection burden during the first year of life were characterized by NMR [ 37 ], demonstrating that infants with a higher infection burden had proinflammatory and proatherogenic plasma metabolomic and lipidomic profiles that in adults suggest an enhanced risk of CVD, obesity, and type 2 diabetes. Therefore, the study of the cardiometabolic associations with common infections and inflammation in early life is very important to perform a risk stratification and develop targeted interventions to prevent CVD implications.…”

“…The only requirement for inclusion at enrolment was the geographical location in which the mother resided and the expected date of delivery (April 1991-Dec 1992). The participants recruited to the study were broadly representative of the general population of new parents' resident in the area at the time in terms of sex, ethnicity and socio-economic status 9 . A significant strength of this study is that we have been able to obtain inter-generational proteomic data from mothers and their offspring, with a large family overlap.…”

“…Inflammation is therefore hypothesised to mediate the negative health effects of a variety of exposures associated with increased inflammation. For example, current evidence indicates that infection and inflammation in early life has important consequences for cardiovascular health later in life 9 . Further, increased levels of inflammation have been reported in adults who had experienced adverse childhood experiences (ACEs) decades earlier 10 possibly explaining why these individuals also experience increased risk of non-communicable diseases.…”
