Early Indicators of Creatine Transporter Deficiency

Abstract: Early identification is a goal for Creatine Transporter Deficiency (CTD), and will be critical for future treatment. Before the first birthday, half of this sample showed both a significant feeding/weight gain issue, and delayed sitting or crawling. Combined, these early indicators could have alerted providers to conduct a urine screen.

“…We observed this difference already after weaning, but taking into account the degree of significance, it is likely that it extends from previous time points. This body weight gain issue is in agreement with reported early signs of CTD 39 .…”

“…These negative results in learning and memory performance contrast with the data from the ubiquitous CrT KO mice with exons 2–4 and 5–7 deleted, where deficits were present although with milder phenotype in NOR test around the same age in the last one 13 , 16 . Interestingly, these results showing normal recognition of an object or a location in KI males are aligned with data from CTD patients, as they are able to remember people, objects or locations but fail in the performance of more complex daily tasks and learning new abilities 17 , 39 . Performance of tasks with little or high level of complexity/difficulty can also be influenced by emotional learning and memory, and by other cognitive processes such as language or attention.…”

“…Despite a precocious onset of symptoms, the average age of diagnosis of CTD ranges from 2 to 20 years [72]. The identification of early clinical indicators, alerting pediatricians to seek a specialty referral and/or conduct a urine screen, might accelerate the diagnosis.…”
