Early Growth Response Factor 1 in Aging Hematopoietic Stem Cells and Leukemia

Abstract: Aging is associated with various hematological disorders and a higher risk of myeloproliferative disorders. An aged hematopoietic system can be characterized by decreased immune function and increased myeloid cell production. Hematopoietic stem cells (HSCs) regulate the production of blood cells throughout life. The self-renewal and regenerative potential of HSCs determine the quality and quantity of the peripheral blood cells. External signals from the microenvironment under different conditions determine the… Show more

“…Analysis of DEGs revealed a reduction in Egr1, an immediate early gene and well known transcriptional target of Srf, in Srf cKO hair cells. Various studies have established Egr1 as a regulator of hematopoietic cell differentiation and maturation (Kulkarni, 2022). Ctgf which was also significantly reduced in Srf cKO hair cells, was found to be enriched in cell growth-associated GO terms.…”

“…Others are produced in the kidney (erythropoietin; thrombopoietin) and in the liver (thrombopoietin). Of special interest is the recently described early response growth factor 1 (ERGF1), 39 which favors the commitment of the HSC to the pluripotent myeloid progenitor and may be responsible for the decreased production of lymphocytes and increased production of granulocytes in the aged population.…”

“…In cell cycle regulation, the transcription factors play a central role by monitoring the cell cycle progression. Early growth response 1 (Egr1) is an essential transcription factor in the maintenance of HSC number and their retention in the BM (Kulkarni, 2022). Min et al (2008) performed Egr1 knockout studies in mice to understand its role in normal hematopoiesis.…”
