Early detection of grapevine leafroll virus in Vitis vinifera using in vitro micrografting

Abstract: Micrografting of grapevine was investigated for its use as a tool in virus indexing of grapevine stock. Cabernet franc and Cabernet sauvignon scions infected with grapevine leafroll-associated closterovirus III (GLRaVIII) were grafted on to virus-free indicator rootstocks of LN 33 and Cabernet sauvignon growing in tissue culture. The two rootstocks and two scions were grafted in all four possible combinations along with two control grafts (virus-free scion on virus-free rootstock). A modified MS Murashige and … Show more

“…Until now, micrografting has been widely used for elimination of various plant pathogens such as viruses from citrus (Murashige et al ., ; Roistacher et al ., ; Juárez et al ., ; Sanabam et al ., ), apple (Huang & Millikan, ), peach (Jonard et al ., ; Conejero et al ., ), plum (Conejero et al ., ) and sweet pepper (Katoh et al ., ), viroids from citrus (Roistacher et al ., ), peach (Barba et al ., ) and avocado (Suarez et al ., ), and phytoplasma from jujuba (Wang et al ., ). This technique has also been applied to the studies of biology of grafting (Pina & Errea, ; Gökbayrak et al ., ; Melnyk et al ., ), cell signalling (Turnbull et al ., ; Yin et al ., ), rejuvenation (Revilla et al ., ), plant regeneration following micropropagation (Onay et al ., ; Badalamenti et al ., ), genetic transformation (Almeida et al ., ; Juárez et al ., ) and cryopreservation (Volk et al ., ; Wang et al ., ), in vitro breeding (Bisognin et al ., ), and in vitro biological indexing of viruses and viroids (Tanne et al ., ; Zilkah et al ., ; Pathirana & Mckenzie, ; Kapari‐Isaia et al ., ; Wang & Valkonen, ). Applications of micrografting to crop improvement have been extensively reviewed by Jonard et al .…”

“…This kind of reverse-cleft micrografting technique is a quite good solution for micrografting when rootstock is very thin in Chinese jujube and maybe in other plants. Pathirana and McKenzie (2005) compared the effect of two scion genotypes on two rootstocks in a grapevine micrografting experiment. There were no significant differences between the success rates for the different combinations of rootstock and scion, and the success of the grafts relied completely on how carefully to place the scion on the cleft of the rootstock.…”

“…Sürgün ucu aşılamanın bazı turunçgillerde farklı anaç-kalem kombinasyonları kullanılarak yapıldığı bildirilmiştir (Edriss and Burger, 1984;Kapari-Isaia et al, 2002). Şaraplık üzüm çeşitlerinde anaç ve çeşitlerde yapılan farklı kombinasyonlarda in vitro aşılamada fazla bir farklılık görülmezken, in vivo yapılan aşılamalarda büyük farklılıkların olduğu görülmüştür (Pathirana and McKenzie, 2005).…”
