Early cleavage of preimplantation embryos is regulated by tRNAGln-TTG–derived small RNAs present in mature spermatozoa

Abstract: tRNA-derived small RNAs (tsRNAs) from spermatozoa could act as acquired epigenetic factors and contribute to offspring phenotypes. However, the roles of specific tsRNAs in early embryo development remain to be elucidated. Here, using pigs as a research model, we probed the tsRNA dynamics during spermatogenesis and sperm maturation, and demonstrate the delivery of tsRNAs from semen-derived exosomes to spermatozoa. By microinjection of antisense sequences into in vitro fertilized oocytes and subsequent single-ce… Show more

“…Of note, tRF5s and i-tRFs are the most enriched tsRNA subpopulation in clusters 10–12. Likewise, tRNA-Gln-TTG, which is enriched exclusively in cluster 11, have been shown to participate in the early cleavage of porcine preimplantation embryos [ 76 ] and injection of tRNA-Gly-GCC fragments into zygotes results in slowdown of embryo development [ 30 ].…”

“…Processing of the collected semen is of utmost importance since it can surely mix up the different allocations of the small RNAs being detected. Many small RNAs have been explored and detected in the male genital tract and semen, including microRNAs (miRNAs), PIWI-interacting RNAs (piRNAs), YRNA and transfer RNA (tRNA)-derived small RNAs (tsRNAs) [ 136 , 182 , 183 , 184 ]. The most commonly explored small RNAs are the single-stranded 22–24 nucleotides miRNAs, considered important regulators of post-transcriptional gene expression [ 182 , 185 ] and detected in the testicles, spermatozoa, free in the SP and within SP EVs [ 181 ].…”

“…Of note, sncRNA in the range 18-26 nt also contain short piRNAs, rsRNAs and tsRNA, which may also be involved in the observed phenotype. In this respect, some tsRNA peaking at epididymis cauda or ejaculated sperm in cattle have also been shown to contribute to early cleavage of porcine preimplantation embryos (Chen et al 2020) and injection of tRNA-Gly-GCC fragments into zygotes results in slowdown of embryo development (Sharma et al 2016).…”
